不同配伍比例黄芪-白术水煎剂中黄芪黄酮溶出规律的研究

摘要 本研究旨在通过HPLC法测定黄芪-白术水煎剂中4种黄芪黄酮类的含量,探讨黄芪配伍不同比例白术后黄酮类活性成分的溶出规律。试验方法:采用Dikma Diamonsil Plus C18色谱柱(250mm×4.6 mm,5μm);流动相为0.1%甲酸水溶液-乙腈,梯度洗脱;流速1 mL·min-1;检测波长254 nm;柱温25℃;进样量10μL。结果表明:4种黄芪黄酮类成分的溶出率为毛蕊异黄酮苷>毛蕊异黄酮>芒柄花苷≈刺芒柄花素,在所观察的5个黄芪-白术配伍质量比例(1∶0,2∶1,1∶1,2∶3,1∶2)中,毛蕊异黄酮苷、毛蕊异黄酮和刺芒柄花素的溶出率随着黄芪-白术配伍比例增加而整体呈下降趋势,芒柄花苷的溶出率则呈先上升后下降的趋势,在黄芪-白术1∶1时达到最大值。由此可见,黄芪-白术的配伍比例会影响黄芪黄酮类成分在水煎剂中的溶出率。 This research aims to establish HPLC method for the determination of four Astragalus Flavonoids in Astragalus Membranaceus-Atractylodes Macrocephala water decoction,and to explore the principle of flavonoid dissolution after different proportion of Astragalus Membranaceus was mixed with Atractylodes Macrocephala.The separation was performed on Dikma Diamonsil Plus C18(250 mm×4.6 mm,5μm)with mobile phase composed of 0.1%formic acid-acetonitrile solution for gradient elution at the flow rate of 1 mL·min^(-1),the detection wavelength was set at 254 nm,the column temperature was set at 25℃,and the injection volume was 10μL.The order of flavonoid dissolution rate from high to low was Calycosin-7-glucoside,calycosin,ononin,and formononetin.As the Astragalus Membranaceus-Atractylodes Macrocephala compatibility proportions(1∶0,2∶1,1∶1,2∶3,1∶2)increased,the dissolution rate of Calycosin-7-glucoside,calycosin,and formononetin decreased,while the dissolution rate of ononin first increased and then decreased,reaching maximum at the proportion of 1∶1.These results show that the Astragalus Membranaceus-Atractylodes Macrocephala compatibility proportions would affect the Astragalus flavonoid dissolution in water decoction.
