摘要
古菌作为细菌和真核生物之外的第三大生命域,广泛分布于陆生、水生甚至肠道等环境中。迄今为止,有关其病毒的研究依然十分薄弱。通过深度分析滴水湖宏病毒组数据,并结合tRNA基因匹配的方法,对其中的古菌病毒进行了探索。结果表明,发现了一条沃斯古菌门(Candidatu sWoesearchaeota)古菌病毒基因组序列,长度为23340bp,其tRNA基因序列与宿主的具有100%的一致性,将之命名为Candidatus Woesearchaeota Phage DSL-JZ1,简称DSL-JZ1。基因组注释结果显示,DSL-JZ1除了编码多个沃斯古菌的同源蛋白,还包括尾钉蛋白(tail spikeprotein)、尾丝蛋白(tailfiberprotein)以及裂解酶等有尾病毒目病毒结构和毒力基因。基因组共享基因网络分析证明,DSL-JZ1与目前已知的所有古菌病毒间,包括头尾结构的古菌病毒,缺乏明显的亲缘性。结果揭示DSL-JZ1代表着一个全新的、感染沃斯古菌门古菌的、具有头尾结构的、源自我国淡水湖泊的水生古菌病毒。
Archaea,as the third major domain of life comprising prokaryotes and eukaryotes,and widely distributed in terrestrial,aquatic,and even intestinal environments.To date,studies on archaeal viruses are still very weak.In this study,we explored the archaeal viruses in Dishui Lake by analyzing its metaviromic data with the method of tRNA gene matching.The results showed that a virus genome of Candidatus Woesearchaeota was identified with a length of 23 340 bp,and its tRNA gene is 100% identical to that of the host.The virus was named Candidatus Woesearchaeota phage DSL-JZ1(DSL-JZ1).Functional annotation of genes indicated that DSL-JZ1 encodes proteins homologous to that of Candidatus Woesearchaeota as well as of structural and virulent proteins of tail spike protein,tail fiber protein,and lyase from caudoviruses.Genome-shared gene network analysis demonstrated that DSL-JZ1 barely bears affinity with all known archaeal viruses,including archaeal viruses with head-to-tail structures.The results demonstrated that DSL-JZ1 represents a novel aquatic archaeal virus with head-to-tail structure and infects the Candidatus Woesearchaeota host originating from a freshwater lake in China.
