Determination of Tetrahydrocannabinol in Food by Colloidal Gold Immunochromatographic Assay

摘要 本研究建立了一种检测食品中四氢大麻酚胶体金免疫层析法。利用胶体金标记四氢大麻酚单克隆抗体,研制大麻胶体金免疫层析试纸条及微孔冻干金标抗体,验证试纸条的灵敏度、特异性、稳定性,对食品中四氢大麻酚进行样品前处理及加标检测,确定了胶体金最适pH值、最适抗体标记量、二抗及抗原包被浓度。在饮料和饼干中,大麻胶体金免疫层析试纸条的检出限为1μg/mL(μg/g)。该试纸条与吗啡、氯胺酮、地西泮、可卡因、苯巴比妥、美沙酮无交叉反应,特异性良好,可测得37℃下14 d良好稳定性。本方法能快速检测食品中非法添加的四氢大麻酚成分,为违禁大麻食品的监管提供理论和技术支持。 A method for rapid determination of tetrahydrocannabinol in food by colloidal gold immunochromatographic assay was established.Colloidal gold was used to label tetrahydrocannabinol monoclonal antibody,and then tetrahydrocannabinol colloidal gold immunochromatographic test strip and micropore freeze-dried gold labeled antibody was developed.The sensitivity,specificity,and stability of test strip were verified.The samples of tetrahydrocannabinol in food were tested after marking and pretreating.The optimal pH value of colloidal gold,the optimal amount of antibody labeling,secondary antibody and antigen coating concentration were determined.In beverages and biscuits,the detection limit of tetrahydrocannabinol colloidal gold immunochromatographic test strip was 1μg/mL(μug/g).The test strip had no cross-reaction with morphine,ketamine,diazepam,cocaine,phenobarbital and methadone,with good specificity.The test strip could be measured at 37C for 14 days with good stability.This method can quickly detect the illegally added tetrahydrocannabinol components in food,and provide theoretical and technical support for the control of illegal cannabis foods.
