桃金娘多酚的响应面优化提取及抗氧化活性研究

Optimization of Extraction and Antioxidant Activity of Polyphenols from Rhodomyrtus tomentosa by Response Surface Methodology

摘要 采用响应面法对桃金娘多酚提取条件进行优化,研究乙醇体积分数、提取温度、料液比、提取时间等因素对桃金娘多酚提取效果的影响,并对桃金娘多酚进行体外抗氧化活性试验。结果表明,响应面优化最优提取工艺为乙醇体积分数45%、提取温度76℃、料液比1︰95 (g/mL),此工艺下可实现32.58±0.24 mg GAE/g的最高多酚提取量。此外,试验发现桃金娘多酚具备较强抗氧化功能,其清除DPPH自由基、ABTS^(+)·自由基的IC_(50)(半抑制浓度)分别是4.47μg/mL和10.93μg/mL。 The response surface methodology was used to optimize the condition of polyphenols extraction from Rhodomyrtus tomentosa.The experiment explored the effects of ethanol concentration,extraction temperature,solid-to-liquid ratio and extraction time on the extraction rate of polyphenols.The results showed that the optimal condition for extraction that provided the maximum yield of polyphenols of 32.58±0.24 mg GAE/g berries was determined as follows:45%ethanol as the extraction solvent,a solid-to-liquid ratio of 1:95(g/mL),an extraction temperature of 76℃.The test of antioxidant activity showed that the polyphenols from Rhodomyrtus tomentosa had better antioxidant activity,and half inhibition concentrations(IC_(50))of ABTS^(+)and DPPH were 10.93μg/mL and 4.47μg/mL,respectively.
