摘要
目的:探讨柴胡皂苷d对卵清蛋白(OVA)诱导的过敏性哮喘小鼠p38丝裂原活化蛋白激酶/核因子κB(p38 MAPK/NF-κB)信号通路及气道炎症的影响。方法:60只BALBc小鼠随机分为正常组、模型组、柴胡皂苷d低(25 mg/kg)、中(50 mg/kg)、高剂量组(100 mg/kg)、阳性对照组(地塞米松,10 mg/kg),每组10只。蛋白免疫印迹法检测小鼠肺组织中p38 MAPK/NF-κB通路相关蛋白的表达。结果:与模型组相比,柴胡皂苷d低、中、高剂量组、阳性对照组小鼠气道阻力参数Penh值、肺泡灌洗液中炎性因子水平、炎性细胞数量、肺组织炎症细胞浸润、肺组织中p38 MAPK/NF-κB通路相关蛋白表达降低(P<0.05)。结论:柴胡皂苷d可减轻OVA诱导的过敏性哮喘小鼠的气道炎症反应,其机制可能与抑制p38 MAPK/NF-κB通路有关。
Objective: To investigate the effects of saikosaponin d on p38 mitogen-activated protein kinase/nuclear factor-κB(p38 MAPK/NF-κB) signaling pathway and airway inflammation in mice with allergic asthma induced by ovalbumin(OVA). Methods: Sixty BALBc mice were randomly divided into normal group, model group, saikosaponin d low-dose(25 mg/kg), medium-dose(50 mg/kg), and high-dose groups(100 mg/kg), and positive control group(dexamethasone, 10 mg/kg), with 10 rats in each group. The expression of p38 MAPK/NF-κB pathway related proteins in mouse lung tissue was detected with Western Blot. Results: Compared with that respectively in the model group, the Penh value, the level of inflammatory factors in the alveolar lavage fluid, the number of inflammatory cells, the infiltration of inflammatory cells in the lung tissue, and the expression of p38 MAPK/NF-κB pathway related proteins in the saikosaponin d low-, medium-, and high-dose groups and positive control group was significantly reduced(P<0. 05). Conclusion: Saikosaponin d can alleviate airway inflammation in mice with allergic asthma induced by OVA, and its mechanism may be related to the inhibition of p38 MAPK/NF-κB pathway.
