摘要
目的探讨坚骨胶囊对骨质疏松模型大鼠骨密度(BMD)、生物力学特性,以及血清细胞因子、股骨组织蛋白表达的影响。方法将60只Wistar雌性大鼠随机分为假手术组(A组,等体积生理盐水),模型组(B组,等体积生理盐水),仙灵骨葆组[C组,468 mg/(kg·d)]及坚骨胶囊高、中、低剂量组[D_(1)组、D_(2)组、D_(3)组,2184,1092,546 mg/(kg·d)],各10只。A组大鼠仅切除卵巢周围少许脂肪组织,其余组大鼠行双侧卵巢切除术,以复制骨质疏松大鼠模型。建模6周后各组大鼠灌胃相应药物或生理盐水,每日1次,连续8周。观察大鼠股骨BMD及股骨生物力学特性(最大载荷、骨应力、结构强度)的变化;采用酶联免疫吸附(ELISA)法检测大鼠血清雌二醇(E_(2))、骨碱性磷酸酶(BALP)、骨钙素(BGP)水平;采用Western blot法检测股骨组织中Wnt、β-catenin、淋巴增强因子(LEF)、骨形成蛋白2(BMP-2)蛋白表达水平。结果与B组比较,D_(1)组、D_(2)组大鼠股骨BMD值均显著升高(P<0.05或P<0.01);D_(1)组大鼠股骨最大载荷、骨应力及结构强度均显著升高(P<0.05或P<0.01),D_(2)组大鼠股骨最大载荷、骨应力均显著升高(P<0.05);D_(1)组、D_(2)组大鼠血清E_(2)含量均显著升高,BALP及BGP含量均显著降低(P<0.05或P<0.01);D_(1)组、D_(2)组大鼠股骨组织中Wnt、β-catenin、LEF蛋白及D_(1)组大鼠股骨组织中BMP-2蛋白的表达水平均显著升高(P<0.01)。结论坚骨胶囊可改善骨质疏松模型大鼠BMD及生物力学特性,其机制可能与影响Wnt/β-catenin信号通路有关。
Objective To investigate the effect of Jiangu Capsules on bone mineral density(BMD),biomechanical properties,serum cytokines,and protein expression in femoral tissue in osteoporosis model rats.Methods Sixty female Wistar rats were randomly divided into the sham-operation group(group A,equal volume of normal saline),model group(group B,equal volume of normal saline),Xianlinggubao group[group C,468 mg/(kg·d)],and high-,medium-,and low-dose groups of Jiangu Capsules[groups D_(1),D_(2) and D_(3),2184,1092,546 mg/(kg·d)],with 10 rats in each group.In group A,only a small amount of adipose tissue around the ovaries was removed from the rats,while in the other groups,bilateral ovariectomy was performed to replicate the osteoporosis rat model.After six weeks of modeling,rats in each group were given corresponding drugs or normal saline by gavage once a day for eight weeks.The change of BMD and biomechanical properties(maximum load,bone stress,structural strength)of the femur in rats was observed.The serum estradiol(E_(2))levels,bone alkaline phosphatase(BALP)levels,and osteocalcin(BGP)levels of rats were measured by the enzyme-linked immunosorbent assay(ELISA).The expression levels of Wnt,β-catenin,lymphocyte enhancement factor(LEF),and bone morphogenetic protein 2(BMP-2)protein were detected by the Western blot.Results Compared with those in group B,the BMD of the femurs in groups D_(1) and D_(2) were significantly higher(P<0.05 or P<0.01);the maximum load,bone stress,and structural strength of the femur in group D_(1) were significantly higher(P<0.05 or P<0.01),and the maximum load and bone stress of the femur in group D_(2) were significantly higher(P<0.05);the serum E_(2) content in groups D_(1) and D_(2) was significantly higher,while the BALP and BGP content in groups D_(1) and D_(2) was significantly lower(P<0.05 or P<0.01);the expression levels of Wnt,β-catenin,and LEF protein in the femoral tissue in groups D_(1) and D_(2) and the expression level of BMP-2 protein in the femoral tissue in group D_(1) were si
