摘要
近年来,莼菜叶腐病在湖北利川莼菜产区大量发生,导致莼菜产量与质量下降,严重制约了当地莼菜产业的发展。本研究旨在明确莼菜叶腐病病原菌旋柄腐霉(Phytopythium helicoides)的生物学特性,为该病的防治提供理论依据。以旋柄腐霉为试验材料,采用菌丝生长速率法研究了不同培养基、pH值、温度、光照条件、碳氮条件对该病原菌菌丝生长速率的影响。结果表明,最适合该菌生长的培养基为V8培养基,其次为PSA与PDA培养基;最适生长pH值为6、最适生长温度为35℃、在12 h光暗交替条件下菌丝生长速度最快、最适碳源氮源分别为蔗糖与甘氨酸。该研究结果可为莼菜叶腐病发病规律研究及防治提供科学依据。
In recent years,leaf rot of Brasenia schreberi has occurred in large numbers of areas in Lichuan,resulting in a decline in the yield and quality of B.schreberi,which seriously restricted the development of the local lettuce industry.The purpose of this study was to clarify the biological characteristics of Phytopythium helicoides,a pathogen of lettuce leaf rot,and to provide a theoretical basis for the prevention and treatment of this disease.The effects of different media,pH,temperature,light conditions,carbon and nitrogen conditions on the growth rate of pathogenic mycelium were studied by mycelium growth rate method.The results showed that the most suitable medium for the growth of this bacterium was V8 medium,followed by PSA and PDA medium.The optimal growth pH value was 6,the optimal growth temperature was 35℃,the mycelium growth rate was the fastest under the condition of alternating light and dark at 12 h,and the most suitable carbon source and nitrogen source were sucrose and glycine.The results of this study could provide scientific basis for the study and prevention of leaf rot of B.schreberi.
