摘要
樟具有多种化学型,以其所含的主要成分(萜类)作为划分依据。为探讨龙脑型和樟脑型樟所含萜类的关键基因和代谢通路,对龙脑型樟和樟脑型樟叶片进行转录组和代谢组测序。转录组分析发现,两组样本间存在差异表达基因2345个,其中上调表达的有882个,下调的有1553个。代谢组共鉴定了468种差异代谢物,上、下调的差异代谢物分别为249种和219种。在单萜的生物合成途径中,发现差异基因9个,差异代谢物8个。在萜类骨架合成中,MEP和MVA均可合成单萜通用底物。其中,龙脑樟在MEP途径高通量表达且限速酶DXS基因表达量显著上升。结合转录组和代谢组可知,龙脑樟趋向于通过MEP途径合成单萜通用底物并进一步生成各类单萜。
Cinnamomum camphora has a variety of chemical types,and its main components(terpenoids)are used as the basis for classification.In order to investigate the key genes and metabolic pathways of terpenoids contained in camphor-type and borneol-type,transcriptomics and metabonomics were performed on the leaves.Transcriptomics analysis showed that there were 2345 differentially expressed genes between the two groups,among which 882 were up-regulated and 1553 were down-regulated.A total of 468 differential metabolites were identified in the metabonomics,with 249 and 219 down-regulated differential metabolites,respectively.In the monoterpene biosynthetic pathway,9 differential genes and 8 differential metabolites were found.In the synthesis of terpenoid skeleton,both MEP and MVA could synthesize monoterpenoid substrates,of which the MEP pathway showed a high throughput expression of borneol-type and the quantity of expression of DXS gene significantly increased.Combined with the ranscriptomics and metabonomics,it was found that borneol-type camphor tended to synthesize monoterpenoid substrates through the MEP pathway and then further produce various monoterpenoids.
