基于生物信息学探讨miRNA在糖尿病合并冠心病患者中的意义 被引量:2

SIGNIFICANCE OF MIRNA WAS EXPLORED IN DIABETES WITH CORONARY HEART DISEASE BASED ON BIOINFORMATICS

摘要 目的 基于生物信息学探讨影响2型糖尿病(T2DM)合并冠状动脉心脏病(CHD)的潜在机制,通过筛选T2DM合并与不合并CHD患者中差异miRNA,观察差异miRNA在T2DM合并CHD中的作用及临床价值。方法 选取GEO数据库中GSE185845数据集并经GEO2R在线平台筛选差异miRNA,并经TargetScanHuman网站预测调控的基因,绘制火山图、PPI图和KEGG富集图并进行相应的分析。结果 从GSE185845数据集中筛选出2个差异miRNA,分别为miR-4465和miR-6089,相关分析提示miR-4465和miR-6089呈正相关(r=0.577,P <0.01)。PPI图显示与差异miRNA共有的基因有2个,为SNN和GMFB,主要富集于MAPK靶点相关的信号通路、FCGR3A调控的磷酸化、固有免疫系统等。生物过程主要富集于Arp2/3复合物调控的肌动蛋白核酸、同源修复基因缺陷等;分子功能主要富集于肌动蛋白结合、NO的激活、细胞色素B5还原酶的激活等;细胞组分主要富集于肌动蛋白细胞骨架,Arp2/3蛋白复合物和细胞与细胞的交联等。结论 miR-4465和miR-6089可能是导致T2DM并发CHD的潜在靶miRNA,这可能与其调控的SNN和GMFB表达有关,机制可能涉及到MAPK靶点相关的信号通路。 Objective To explore the potential mechanism of type2 Diabetes(T2DM) with Coronary Heart Disease(CHD) using bioinformatics,and to observe the clinical significance of differential miRNA,which was screened between T2DM with CHD and without CHD patients.Methods GSE185845 datasets were selected from GEO database.The differential miRNA was screened through GEO2R online platform,and gene was regulated and predicted from TargetScanHuman website,volcano graph graph,PPI network diagram,KEGG enrichment plot were drawn and analyzed.Results 2 differential miRNAs were screened from GSE185845 dataset,miR-4465 and miR-6089 respectively,which is positive connected with each other(r=0.577,P<0.01).2 genes co-regulated by differential miRNA were showed in PPI diagram,MAPK target-related signaling pathways,AMPK signaling pathway,FCGR3A-regulated phosphorylation and innate immune system and so on were mainly enriched.Arp2/3 complex-mediated actin,break repair via homologous and so on are concerned in biological process;Actin binding,nitrite reductase activity,cytochrome-b5-reductase activity acting on NADPH and so on are enriched in molecular function;Actin cytoskeleton,Arp2/3 protein complex and cell-cell junction and so on are concerned in cellular component.Conclusion miR-4465 and miR-6089 maybe potential target miRNA of T2DM with CHD,which may be associated with SNN and GMFB expressions regulated by miR-4465 and miR-6089,The mechanism may be connected with MAPK target-related signaling pathways.
