乙醇/硫酸铵双水相体系萃取纯化紫甘薯色素 被引量:1

Purification of Purple Potato Pigment by Ethanol/Ammonium Sulfate Aqueous Two-Phase System

摘要 为了除去紫甘薯提取液中的糖类杂质,采用乙醇/硫酸铵双水相体系纯化紫甘薯色素提取液。通过设计ω(乙醇)、ω(硫酸铵)、色素提取液pH以及ω(色素提取液)四个因素的单因素实验和正交实验来探究和优化实验条件。通过正交实验,以选择性系数为考察指标,确定了各因素对紫甘薯色素纯化的影响程度为:ω(乙醇)>ω(硫酸铵)>pH>ω(色素提取液);最优纯化条件是:ω(乙醇)为25.0%,ω(硫酸铵)为17.0%,色素提取液pH为3.5,ω(色素提取液)为11.0%,此时的选择性系数α为242.07,色素保留率为98.88%,糖类去除率为85.25%。该方法分离纯化紫甘薯色素产品易回收、生产成本低,可提高企业的经济效益,有望后续进行工业化生产。 In order to remove the saccharide impurities in purple sweet potato extract,aqueous two-phase system of ethanol-ammonium sulfate is used to purify the extract.The single factor experiments and orthogonal experiments based on ethanol,ammonium sulfate,pigment extract and pH,are investigated to explore and optimize the process conditions.The influence degree of each factor on the purification of purple sweet potato pigment are determined by the orthogonal experiment with the selectivity coefficient(α)as an index:ethanol mass fraction>ammonium sulfate mass fraction>pH>pigment extract addition amount.The optimum parameters are as follows:ethanol 25.0%,ammonium sulfate 17%,pH 3.5,and pigment extract 11.0%,resulting in a selectivity coefficient of 242.07,a pigment retention rate of 98.88%and a sugar removal rate of 85.25%.The results indicate that this method can effectively separate and purify purple sweet potato pigment with easy recovery of product and low production cost,which can improve the economic benefits of enterprise,and is promising for subsequent industrial production.
