用18S rRNA高通量测序分析比较万峰湖丰水期和枯水期生物多样性 被引量:1

Comparison of Biodiversity Between Wet Season and Dry Season in Wanfeng Lake by 18S rRNA High Throughput Sequencing

摘要 为了比较贵州省黔西南州万峰湖丰水期和枯水期真核浮游生物的多样性,分别于2020年丰水期(11月)和2021年枯水期(5月)对兴义市万峰湖进行2次共计20个地点进行采样,采用18S r RNA高通量测序技术研究其群落组成。结果表明:序列多样性和丰度分析显示,丰水期OTUs平均为325个,枯水期OTUs平均198个,丰水期物种的丰度比枯水期丰富,而群落分布多样性和物种均匀度均低于枯水期。通过两次调查,丰水期真核浮游生物主要由节肢动物门、绿藻门和壶菌门等23个门,枯水期主要由绿藻门、节肢动物门、轮虫动物门等16门的生物组成。其中,丰水期的优势物种为节肢动物门,枯水期优势物种为绿藻门。在种的水平上,丰水期由147种生物组成,枯水期由187种生物组成,两个时期均有的物种有马索隐藻、拟蛋白核隐藻和光滑鱼鳞藻等,丰水期的优势物种为右突新镖水蚤、甲藻和马索隐藻;枯水期的优势物种为真镖水蚤、马索隐藻和拟二叉角甲藻。研究表明,万峰湖丰水期和枯水期真核浮游生物群落组成和多样性存在显著差异。通过分析浮游生物群落、优势种、浮游生物丰度、多样性指数和均匀度指数分析,评价万峰湖的水质状况,为研究喀斯特水体中生物多样性奠定基础。 In order to compare the diversity of eukaryotic plankton in the wet season and dry season of Wanfeng Lake in Qianxian Prefecture, Guizhou Province, water samples were collected in Wanfeng Lake in Xingyi City in November 2020(wet season)and May 2021(dry season), and the community composition was studied by 18S r RNA high-throughput sequencing technology. The results showed that the average number of OTUs in the wet season was 325 and 198 OTUs in the dry season, and the abundance of species in the wet season was richer than that in the dry season, while the community distribution diversity and species uniformity were lower than those in the dry season. Through investigation, eukaryotic plankton in the wet season mainly consists of 23 phylums such as Arthropods, Green algae and Chytrid, and the dry season is mainly composed of 16 phylums such as Green algae, Arthropods and Rotifers. Among them, the dominant phylums in the wet season is the Arthropoda, and the dominant phylums in the dry season is Green algae. At the species level, the wet season consists of 147 species, and the dry season consists of 187 species. The common species in both periods are Masolococcus, Cryptococcus pyrenoidosa and Ichthyosquas. The dominant species in the wet season are Neodiaptomus schmackeri, Dinoflagellate and Cryptomonas marssonii and the dominant species in the dry season are Eudiaptomus glaciloides, Cryptomonas marssonii and Ceratium. This study indicated there were significant differences in eukaryotic plankton community composition and diversity in the wet and dry seasons of Wanfeng Lake. Through the analysis of plankton community,dominant species, plankton abundance, diversity index and uniformity index, the water quality status of Wanfeng Lake was evaluated,which laid a foundation for the study of biodiversity in karst water.
