摘要
本试验旨在研究德宏奶水牛SCD基因表达规律和探究水牛SCD生物学特性。用乳品分析仪测定乳脂率,依照乳脂率高低进行分组,分为高、中、低组(分别为H组、M组、L组)。采用qPCR法检测SCD基因表达量,与乳脂率进行关联分析。利用生物信息学方法对水牛SCD的结构进行预测。结果表明,H组SCD表达量极显著高于M组和L组(P<0.01),M组极显著高于L组(P<0.01)。其mRNA表达水平与乳脂率呈强正相关。生物信息学分析结果显示,水牛SCD为不稳定亲水蛋白,不存在信号肽,主要在内质网(44.4%)发挥作用。二级结构主要由无规则卷曲(40.39%)、α-螺旋(39.55%)构成。SCD与ELOVL6、SREBP1、FASN等脂质代谢关键分子互作。研究表明,德宏奶水牛SCD基因的表达可能影响乳脂合成。
The aim of experiment was to study the expression of stearyl-CoA desaturase(SCD)gene and the biological characteristics of SCD protein in Dehong dairy buffalo.Milk fat percentage was detected by milk composition analyzer.High,medium and low groups(H group,M group and L group)were divided by different milk fat content.The qPCR was utilized to detect the expression of SCD gene,and the correlation analysis was utilized for the expression and the milk fat content.The structural characteristics of SCD protein of buffalo was analyzed by bioinformatics method.The results demonstrated that the mRNA expression of SCD gene in group H was extremely significantly higher than that in group M and group L(P<0.01).Group M was extremely significantly higher than group L(P<0.01).The mRNA expression level of SCD gene was an strongly and positively correlated with milk fat percentage.The bioinformatics analysis results showed that SCD was an unstable hydrophilicity protein,and it was not a signal peptide.It was mainly in the endoplasmic reticulum(44.4%).The secondary structure of buffalo was mainly composed of random curl structures(40.39%)andα-helix(39.55%).The SCD had interaction with ELOVL6,SREBP1,and FASN which key molecules of lipid metabolism.The experiment indicated the expression of SCD gene of Dehong dairy buffalo could impact milk fat percentage.
