丹参多酚酸盐对兔急性心肌梗死后心肌凋亡的影响

摘要 目的通过构建新西兰兔的心肌梗死模型,使用不同剂量丹参多酚酸盐静脉输注观察对兔急性心肌梗死后细胞凋亡的影响。方法选取28只健康新西兰兔,构建心肌梗死模型。建模后将心肌梗死兔随机分为4组,分别为对照组、实验组1、实验组2、实验组3,每组7只。实验组1、2、3分别给予1,2,4 mg·kg^(-1)丹参多酚酸盐静脉输液,对照组给予20 mL生理盐水。检测各组兔心功能指标;荧光TUNEL染色观察兔心肌组织中细胞凋亡情况;苏木素-伊红(HE)染色观察兔心肌组织病理学变化;免疫组化检测N钙黏蛋白、肌球蛋白及间隙连接蛋白43的表达;Western blot检测相关蛋白水平。结果与对照组相比,实验组1、2、3兔的EF和LVEDFs水平升高,LVIDd水平降低。实验组1兔的EF和LVEDFs水平最高,LVIDd水平最低,而实验组3兔的EF和LVEDFs水平最低,LVIDd水平最高(P<0.05)。与对照组相比,实验组1、2、3兔的心肌组织中细胞凋亡率均降低,实验组1兔心肌组织中细胞凋亡率低于实验组2,实验组2低于实验组3(P<0.01)。实验组3兔局部心肌层内心肌纤维灶状坏死较为明显,坏死心肌细胞的细胞核深染、固缩,细胞质溶解甚至基本消失。与实验组3相比,实验组1和2兔心肌组织病理症状明显减轻。与对照组相比,实验组1、2、3兔的N-钙黏蛋白、肌球蛋白、间隙连接蛋白43的表达水平均升高,实验组1高于实验组2,实验组2高于实验组3(P<0.05)。心肌细胞中AKT、p-akt、肌动蛋白与IGF-1结合的表达水平均高于心肌细胞中AKT、p-akt、肌动蛋白与IGF-1未结合的表达(P<0.05)。结论使用不同浓度的丹参多酚酸盐对心肌梗死后兔进行治疗,降低了兔心肌细胞的凋亡,为中成药治疗心肌梗死的效果提供依据,也为细胞治疗的临床实际应用提供理论基础。 OBJECTIVE To establish the myocardial infarction model of New Zealand rabbits,and observed the effect of different doses of salvia polyphenol acid on apoptosis of rabbits after acute myocardial infarction by intravenous infusion.METHODS 28 healthy New Zealand rabbits were selected to establish myocardial infarction model.After modeling,rabbits with myocardial infarction were randomly divided into 4 groups:control group,experimental group 1,2 and 3,with 7 rabbits in each group.Experimental group 1,2 and 3 was given 1,2,4 mg·kg^(-1) salvia miltiorrhiza Ⅳ infusion,respectively.Control group was given placebo,20 mL normal saline.Cardiac function indexes were detected in each group.Fluorescence TUNEL staining was used to observe cell apoptosis in myocardial tissue of rabbits.Hematoxylin-eosin(HE)staining was used to observe the pathological changes of myocardial tissue in rabbits.The expressions of N cadherin,myosin and gap junction protein 43 were detected by immunohistochemistry.The protein levels were detected by Western blot.RESULTS Compared with the control group,the levels of EF and LVEDFs in experimental group 1,2,and 3 were increased,and the levels of LVIDd were decreased.The levels of EF and LVEDFs in experimental group 1 were the highest,and the level of LVIDd was the lowest,while the levels of EF and LVEDFs in experimental group 3 were the lowest,and the level of LVIDd was the highest(P<0.05).Compared with the control group,the apoptosis rate of myocardial tissue in experimental group 1,2 and 3 were decreased,and the apoptosis rate of myocardial tissue in experimental group 1 was lower than that in experimental group 2,and that in experimental group 2 was lower than that in experimental group 3(P<0.01).In experimental group 3,focal necrosis of myocardium fibers in the local myocardium was more obvious,and the nuclei of necrotic cardiomyocytes were hyperchromatic,pyknotic,and cytoplasmic lysis was even basically disappeared.Compared with experimental group 3,the pathological symptoms of myocardial tissue
