靶向溶酶体降解在细胞自噬和凋亡中的调控及其异常相关疾病的研究进展

摘要 溶酶体是一种富含多种酶的细胞内消化器,在细胞代谢过程中主要扮演了“清洁工”的角色。当溶酶体降解系统感知外源或某些信号刺激时,可促进其降解酶的释放,发挥降解作用。重要的是,溶酶体在细胞自噬及细胞凋亡中具有重要的调控作用。溶酶体可参与细胞凋亡,并通过自噬降解途径维持细胞内环境稳态。另外,溶酶体功能异常还可诱发多种疾病,如溶酶体贮积症、肿瘤、禽类痛风等。本文以溶酶体的降解途径作为背景,概述其在细胞自噬和细胞凋亡中的调控作用,对其调控异常引起的相关疾病进行综述,为靶向溶酶体的降解技术在相关疾病治疗方面提供部分参考依据。 Lysosomes are intracellular organelle containing a variety of enzymes,and play a vital role of “cleaner” in the cell metabolism.The lysosomal degradation system can promote the release of degradation enzymes when it senses the stimulation of exogenous or some signal,and play an effective role in degradation.Lysosomes are important regulators of apoptosis and autophagy.Importantly,lysosomes have also a critical role in apoptosis and autophagy through involvement of cellular apoptosis and autophagy-mediated degradation pathway,maintaining the intracellular homeostasis.In addition,lysosomal dysfunction also leads to the multiple diseases,such as Lysosomal Storage Disorder(LSD),Tumors,Avian Gout,etc.Therefore,this review aims to summarize the role of lysosomal degradation pathway in the regulation of autophagy and apoptosis and their abnormal regulation-related diseases,and the lysosomal degradation pathway as a research background,to provide reference for the treatment of related diseases with targeted lysosomal degradation technology.
