摘要
[目的]本试验探究药物米非司酮的抗伪狂犬病毒(PRV)作用及其与另一种抗伪狂犬病毒药物氢醌的协同作用,为抗PRV药物的研究和临床应用奠定基础。[方法]用Western blot和qPCR等方法检测米非司酮及其与氢醌联用在体外对伪狂犬病毒复制的影响及其影响的作用阶段,再通过小鼠攻毒保护试验进一步确认其抗病毒作用及与氢醌联用的协同作用。[结果]15μmol·L^(-1)米非司酮具有抗病毒作用,且随着浓度升高而增强,此作用发生于伪狂犬病毒复制早期。体内、体外试验结果表明,米非司酮(体内5μmol·L^(-1)、体外40 mg·kg^(-1))单独或与氢醌(体内5μmol·L^(-1)、体外50 mg·kg^(-1))联用均具有抗病毒作用,与氢醌具有协同抗病毒作用,同时能减轻单独用药对肝脏的损伤。[结论]米非司酮在病毒复制早期发挥抗病毒作用,与氢醌的联用具有更好的抗病毒效果,既减轻氢醌对肝脏的损伤,又弥补米非司酮药效上的不足。
[Objectives]The anti-PRV effect of the drug mifepristone and its synergistic effect with another anti-PRV drug,hydroquinone were investigated to lay the foundation for the research and clinical application of anti-PRV drugs.[Methods]The effect of mifepristone and its combination with hydroquinone on PRV replication in vitro and the stage of action that produced the effect were examined by Western blot and qPCR,and its antiviral effect and synergistic effect with hydroquinone combination were further confirmed by mouse attack protection test.[Results]The antiviral effect of mifepristone at 15μmol·L^(-1)increased with increasing drug concentration and occurred at an early stage of PRV replication.The results of in vitro and in vivo tests showed that mifepristone(5μmol·L^(-1)in vivo,40 mg·kg^(-1) in vitro)alone or in combination with hydroquinone(5μmol·L^(-1)in vivo,50 mg·kg^(-1) in vitro)had antiviral effects and synergistic antiviral effects with hydroquinone.It also reduced the damage to the liver caused by hydroquinone.[Conclusions]Mifepristone had an antiviral effect in the early stage of virus replication,and the combination with hydroquinone had a better antiviral effect.At the same time,it could reduce the damage of hydroquinone to the liver and make up for the efficacy of mifepristone.
