摘要
采用平板组织分离法从土沉香结香部位分离内生真菌,通过沉香主要成分苄基丙酮耐药性初筛及打洞输液法结香复筛土沉香优势结香真菌,并进行分子鉴定。结果表明,从已结香的土沉香木质部中初步分离出内生真菌34株,通过形态学及ITS分子初步鉴定归于7目7科10属,其中镰刀菌属Fusarium为优势属,其次为毛色二孢属Lasiodiplodia。经苄基丙酮耐药性及人工造香复筛,筛选出6种优势促结香真菌,鉴定为尖孢镰刀菌(Fusarium oxysporum)、藤仓镰刀菌(Fusarium fujikuroi)、层出镰刀菌(Fusarium proliferatum)、腐皮镰刀菌Fusarium solani)、假可可毛色二孢(Lasiodiplodia pseudotheobromae、可可毛色二孢(Lasiodiplodia theobromae)。
Endophytic fungi were isolated from agarwood-forming parts of Aquilariae sinensis Resinatum by plate tissue isolation method.The dominant fungi were rescreened and identified by primary screening of benzylacetone resistance and holing infusion method.The results showed that 34 strains of endophytic fungi were isolated from the xylem of Lignum Resinatum.According to the morphological and ITS molecular identification,they were classified into 10 genera,7 families and 7 orders.Fusarium was the dominant genus,followed by Lasiodiplodia.Through the screening of benzylacetone resistance and artificial scenting,6 dominant scenting promoting fungi were screened and identified as Fusarium oxysporum,Fusarium fujikuroi,Fusarium proliferatum,Fusarium solani,Lasiodiplodia pseudotheobromae,Lasiodiplodia theobromae.
