摘要
目的观察温针灸通过调节核因子E2相关因子2(Nrf2)/抗氧化反应元件(ARE)信号通路对卵巢功能减退(DOR)大鼠氧化应激损伤的修复作用。方法将动情周期正常的40只雌性SD大鼠随机分为空白组、模型组、激素组及温针灸组,每组10只。除空白组外,其余组大鼠采用雷公藤多苷片灌胃14 d方法制备DOR模型。造模成功后,激素组给予雌激素替代疗法干预,温针灸组取关元、气海、中极、双侧肾俞进行温针灸干预,均干预14 d。采用阴道涂片观察各组大鼠性周期,ELISA法检测血清中雌二醇(E_(2))、促黄体生成素(LH)、促卵泡激素(FSH)、抗苗勒激素(AMH)、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)、丙二醛(MDA)水平,HE染色观察卵巢组织病理形态,Western blot法及RT-PCR法检测卵巢组织中Nrf2、醌氧化还原酶1(NQO1)、血红素加氧酶-1(HO-1)蛋白和mRNA表达情况。结果与空白组比较,模型组大鼠均出现动情周期紊乱;血清中E_(2)、AMH、SOD、GSH-Px水平均明显降低(P均<0.05),LH、FSH、MDA水平均明显增高(P均<0.05);闭锁卵泡数明显增多(P<0.05),成熟卵泡数明显减少(P<0.05);卵巢组织中Nrf2、NQO1、HO-1蛋白和mRNA表达量均明显降低(P均<0.05)。与模型组比较,温针灸组和激素组大鼠动情周期紊乱比例明显降低(P均<0.05);血清中E_(2)、AMH、SOD、GSH-Px水平均明显升高(P均<0.05),LH、FSH、MDA水平均明显降低(P均<0.05);闭锁卵泡数明显减少(P均<0.05),成熟卵泡数明显增多(P均<0.05);卵巢组织中Nrf2、NQO1、HO-1蛋白和mRNA表达量均明显升高(P均<0.05)。结论温针灸对DOR大鼠具有明显保护作用,其作用机制可能与调控Nrf2/ARE信号通路活化,改善卵巢组织氧化应激状态相关。
Objective It is to observe the repair effect of warm acupuncture on oxidative stress injury in rats with diminished ovarian reserve(DOR)by regulating nuclear factor erythroid 2 related factor 2(Nrf2)/antioxidant response element(ARE)signal pathway.Methods Forty female SD rats with normal estrous cycle were randomly divided into 4 groups:blank group,model group,hormone group and warm acupuncture group,with 10 rats in each group.The rats in each group except for the blank group were treated with Tripterygium wilfordii polyglycoside tablets by gavage for 14 days to establish DOR models.After successful modeling,the hormone group was treated with estrogen replacement therapy,and the warm acupuncture group was treated with warm acupuncture at Guanyuan,Qihai,Zhongji and bilateral Shenshu,all the groups were treated for 14 days.The sexual cycle of rats in each group was observed with vaginal smears,the levels of estradiol(E_(2)),luteinizing hormone(LH),follicle stimulating hormone(FSH),anti Mullerian hormone(AMH),superoxide dismutase(SOD),glutathione peroxidase(GSH-Px)and malondialdehyde(MDA)in serum were determined by ELISA,the pathological morphology of ovaries was observed by HE staining,the expression of protein and mRNA of Nrf2,quinone oxidoreductase 1(NQO1)and heme oxygenase 1(HO-1)in ovarian tissues were detected by Western blot and RT-PCR.Results Compared with the blank group,the estrous cycle of all rats in the model group was disorder;the levels of serum E_(2),AMH,SOD and GSH-Px were significantly decreased(all P<0.05),and the levels of serum LH,FSH and MDA were significantly increased(all P<0.05);the number of atretic follicles was significantly increased(P<0.05),and the number of mature follicles was significantly decreased(P<0.05);the expression of Nrf2,NQO1,HO-1 protein and mRNA in the ovarian tissues were significantly decreased(all P<0.05).Compared with the model group,the proportion of estrous cycle disorder of the rats in the hormone group and warm acupuncture group was significantly lower than that in
