酶协同微波提取云南沙棘果实总黄酮及其抗氧化活性评价 被引量:6

Evaluation of Hippophae rhamnoides fruit total flavonoids extracted by microwave assisted enzymatic method and its antioxidant capacity

摘要 采用纤维素酶协同微波法提取西藏林芝产云南沙棘果实总黄酮。通过Box-Behnken设计对单因素试验筛选出的酶用量、液固比、乙醇体积分数等3个主要提取得率影响因子进行进一步优化,再通过铁离子还原能力(FRAP)和DPPH·、ABTS+自由基清除能力检测,考察了提取物不同极性部位的体外抗氧化能力。结果表明,最优提取参数为:酶用量15 mg/g,液固比50∶1(mL∶g),乙醇体积分数50%,微波功率280 W,提取时间6 min,总黄酮提取得率为31.25 mg/g。质量浓度1.0 g/L时,提取物乙酸乙酯部位的铁离子还原能力最佳,其FRAP值为0.38 mmol/L,该部位对DPPH·、ABTS^(+)自由基的最大清除率也高于其他极性部位,分别为97.65%和80.69%。优化后的协同提取工艺较为稳定,且提取物不同极性部位均具有一定的抗氧化活性,以乙酸乙酯部位活性最佳,可作为天然抗氧化剂开发利用。 The total flavonoids from Yunna Hippophae rhamnoides fruit cultivated at Linzhi,Tibet were extracted by cellulase combined microwave method. Box-Behnken design was used to further optimize extraction conditions.Three main factors which were screened out by single-factor tests were enzyme amount,liquid-solid ratio and ethanol volume fraction. In vitro antioxidant capacity of different polar parts of the flavonoids was evaluated by ferric reducing ability(FRAP)and DPPH·,ABTS^(+)radical scavenging ability. The results showed that the optimal extraction parameters were :enzyme amount 15 mg/g,ratio of liquid to solid 50∶1(mL∶g),ethanol 50%(v/v),microwave power 280 W,and extraction time 6 min. The total extraction yield of flavonoids was 31.25 mg/g. When the concentration was 1.0 g/L,ethyl acetate extract had the best in vitro antioxidant activity,its FRAP value was 0.38 mmol/L,and maximum scavenging rates of DPPH·and ABTS+radical were 97.65% and 80.69%,respectively,which were all higher than those in other polar parts. Comprehensive analysis showed polar extracts all had a certain antioxidant capacity;the ethyl acetate extracts was the best and it can be used as a natural antioxidant in the future.
