摘要
目的:探讨不同检测技术在检测痰液标本中肺炎链球菌(SP)的应用价值。方法:随机选取2018年8月-2020年6月我院收治的获得性肺炎患者50例,运用传统方法、荧光定量PCR法、LAMP法检测所有患者痰液标本中肺炎链球菌,然后统计分析传统方法、荧光定量PCR法、LAMP法检测痰液标本中肺炎链球菌阳性情况,同时统计分析荧光定量PCR法、LAMP法检测痰液标本中肺炎链球菌灵敏度。结果:传统方法检测痰液标本中肺炎链球菌阳性42例,阴性8例,阳性率为84.0%(42/50);荧光定量PCR法检测痰液标本中肺炎链球菌阳性48例,阴性2例,阳性率为96.0%(48/50);LAMP法检测痰液标本中肺炎链球菌阳性49例,阴性1例,阳性率为98.0%(49/50)。荧光定量PCR法、LAMP法检测痰液标本中肺炎链球菌阳性率均显著高于传统方法组(P<0.05),但荧光定量PCR法、LAMP法检测痰液标本中肺炎链球菌阳性率之间的差异不显著(P>0.05)。LAMP法在35 min、60 min时检测痰液标本中肺炎链球菌最低菌液浓度均显著低于荧光定量PCR法(P<0.05),而60 min时最低菌液浓度又显著低于35 min时(P<0.05),灵敏度最高。结论:LAMP法检测痰液标本中肺炎链球菌(SP)的应用价值较传统方法、荧光定量PCR法高。
Objective:To explore the application value of different detection techniques in the detection of Streptococcus pneumoniae(SP)in sputum samples.Methods:50 cases of patients with acquired pneumonia treated in our hospital from August 2018 to June 2020 were randomly selected.Streptococcus pneumoniae in sputum samples of all patients was detected by traditional methods,fluorescence quantitative PCR and lamp methods,and then the positive status of Streptococcus pneumoniae in sputum samples by traditional methods,fluorescence quantitative PCR and lamp methods,Streptococcus pneumoniae in sputum samples sensitivity detect by lamp method were statistically analyzed.Results:42 cases of Streptococcus pneumoniae were positive and 8 cases were negative in sputum samples by traditional methods.The positive rate was 84.0%(42/50);The positive rate of Streptococcus pneumoniae was 96.0%(48/50);In sputum samples,49 cases of Streptococcus pneumoniae were positive and 1 case was negative by lamp method.The positive rate was 98.0%(49/50).The positive rates of Streptococcus pneumoniae in sputum samples detected by fluorescence quantitative PCR and lamp were significantly higher than those in the traditional method group(P<0.05),but there was no significant difference between fluorescence quantitative PCR and lamp(P>0.05).The minimum bacterial concentration of Streptococcus pneumoniae in sputum samples detected by lamp method at 35min and 60min was significantly lower than that detected by fluorescence quantitative PCR(P<0.05),while the minimum bacterial concentration at 60min was significantly lower than that at 35min(P<0.05).Conclusion:The application value of lamp method in the detection of Streptococcus pneumoniae(SP)in sputum samples is higher than that of traditional method and fluorescence quantitative PCR.
