摘要
目的探讨小窝蛋白1(caveolin-1)和表皮生长因子受体(EGFR)在肺腺癌组织的表达及临床意义。方法采用免疫组化法分别检测120例肺腺癌组织及癌周正常肺组织中caveolin-1、EGFR蛋白表达,蝎型探针扩增阻滞突变-荧光定量聚合酶链反应检测肺腺癌组织中EGFR基因突变,并分析其与临床病理因素及预后的关系。结果Caveolin-1蛋白在肺腺癌组织中的阳性表达率低于正常肺组织(53.33%vs.89.17%)(P<0.01);而EGFR蛋白在肺腺癌组织中的阳性表达率高于正常肺组织(68.33%vs.14.17%)(P<0.01)。Caveolin-1蛋白表达与淋巴结转移和TNM分期相关(P<0.05);EGFR蛋白表达与淋巴结转移、分化程度及TNM分期相关(P<0.05);EGFR基因突变与性别构成比、吸烟史相关(P<0.05)。肺腺癌组织中caveolin-1蛋白表达与EGFR蛋白表达、基因突变均呈负相关(rs=-0.280、-0.211,P<0.05)。Caveolin-1和EGFR蛋白阳性表达患者的中位生存时间分别少于阴性表达患者(P<0.01)。结论Caveolin-1和EGFR表达与肺腺癌的进展及患者预后密切相关。Caveolin-1可作为肺腺癌潜在的研究靶点。
Objective To investigate the expressions and clinical significance of caveolin-1 and epithelial growth factor receptor(EGFR)in lung adenocarcinoma tissues.Methods The protein expressions of caveolin-1 and EGFR in 120 cases of lung adenocarcinoma tissues and adjacent normal lung tissues were determined by immunohistochemistry.The gene mutation of EGFR in lung adenocarcinoma tissues was detected by amplifieation refractory mutation system-fluorescent quantitative PCR.The relationship of caveolin-1 and EGFR with the clinical pathological factors and prognosis was analyzed.Results The positive expression rate of caveolin-1 protein in lung adenocarcinoma tissues was lower than that in normal lung tissues(53.33%vs.89.17%)(P<0.01),which of EGFR protein in lung adenocarcinoma tissues was higher than that in normal lung tissues(68.33%vs.14.17%)(P<0.01).The protein expression of caveolin-1 was closely related to the lymph node metastasis and TNM stage(P<0.05).The protein expression of EGFR was closely related to the lymph node metastasis,differentiation degree and TNM stage(P<0.05).The gene mutation of EGFR was associated with gender and smoking history(P<0.05).The protein expression of caveolin-1 was negatively correlated with the protein expression and gene mutation of EGFR in lung adenocarcinoma tissues(rs=-0.280 and-0.211,P<0.05).The median survival time of the patients with positive protein expressions of caveolin-1 and EGFR was less than that of the patients with negative protein expressions,respectively(P<0.01).Conclusion The protein expressions of caveolin-1 and EGFR are closely related with the progression of lung adenocarcinoma and prognosis of the patients.Caveolin-1 may be taken as a potential target for studying lung adenocarcinoma.
