高咬合状态下牙龈组织中VEGF及β-catenin表达的变化及其机制 被引量:1

CHANGES IN VEGF AND BETA-CATENIN EXPRESSION IN GINGIVAL TISSUES UNDER HYPEROCCLUSION AND ITS MECHANISM

摘要 目的通过检测高咬合状态下小鼠牙龈组织中血管内皮细胞生长因子(VEGF)及β-catenin表达的变化,探讨VEGF及Wnt/β-catenin信号通路在牙龈组织改建中的作用。方法选取4周龄昆明种雄性小鼠30只,随机分为对照组和实验组,每组15只小鼠。通过拔除小鼠右侧上颌全部磨牙的方法建立实验组小鼠高咬合模型。分别于术后第3、7、14、28、56天,每组每次随机选取3只小鼠麻醉后处死,分离左侧下颌第一磨牙及牙龈组织,使用苏木精-伊红(HE)染色观察牙龈组织结构的变化,使用免疫组化染色方法检测牙龈组织中VEGF以及β-catenin表达的变化。结果HE染色显示,实验组拔牙后第14天牙龈组织明显增厚,血管数量增多。免疫组化检测结果显示,对照组不同时间点牙龈组织中VEGF、β-catenin表达比较,差异无显著性(P>0.05)。实验组不同时间点牙龈组织中VEGF、β-catenin表达比较,差异有显著性(F=250.4、343.9,P<0.01)。除拔牙后第56天外,两组其余时点牙龈组织VEGF、β-catenin表达比较差异均有显著性(F=5.7~38.4,P<0.05)。结论高咬合状态下,牙龈组织中VEGF及β-catenin表达的变化影响牙龈组织的改建。 Objective To investigate the role of vascular endothelial growth factor(VEGF)and the Wnt/β-catenin signaling pathway in gingival tissues remodeling by measuring the changes in VEGF andβ-catenin expression in mouse gingival tissues under hyperocclusion.Methods Thirty male Kunming mice aged four weeks were randomly divided into control group(n=15)and experimental group(n=15).All the right maxillary molars of mice in the experimental group were extracted to establish a hyperocclusion model.On the 3rd,7th,14th,28th,and 56th days after operation,three mice in each group were randomly selected and sacrificed under anesthesia.Their left mandibular first molars with the gingival tissues were isolated.Hematoxylin-eosin(HE)staining was performed to observe the histological changes of the gingiva,and immunohistochemical staining was used to measure the changes in VEGF andβ-catenin expression in the gingival tissues.Results The HE staining showed that in the experimental group,the thickness of gingival tissues and the number of blood vessels increased on day 14 after tooth extraction.The results of immunohistochemical examination showed that in the control group,there were no significant differences in the expression of VEGF andβ-catenin in the gingival tissues between different time points(P>0.05);while in the experimental group,there were significant differences in that between different time points(F=250.4,343.9,P<0.01).At all time points except the 56th day after tooth extraction,there were significant differences in the expression of VEGF andβ-catenin in the gingival tissues between the two groups(F=5.7-38.4,P<0.05).Conclusion Under hyperocclusion,changes in the expression of VEGF andβ-catenin in gingival tissues affect the remodeling of gingival tissues.
