Simultaneous determination of five flavonoids in leaves of Ampelopsis grossedentata by HPLC-MS/MS

摘要 目的:建立同时测定藤茶中5个黄酮类成分的高效液相色谱串联质谱法。方法:采用CNW Athena C18-WP色谱柱(100 mm×2.1 mm,3μm),以甲醇-0.1%甲酸水溶液为流动相梯度洗脱,流速为0.3 mL·min^(-1),柱温为30℃。采用电喷雾离子源,多反应监测模式,负离子检测。结果:杨梅素、二氢槲皮素、槲皮苷、山柰酚和(-)-表儿茶素没食子酸酯分别在54.4~1030、28.4~1000、27.4~1000、24.8~998、30.6~1010 ng·mL^(-1)范围内线性关系良好(r>0.9968),精密度、稳定性和重复性的RSD均小于5.0%(n=6),平均加样回收率分别为96.4%、99.8%、97.7%、95.0%和100.9%,RSD分别为4.7%、1.4%、1.1%、4.3%和3.0%(n=6)。4批藤茶样品中测得的上述5个成分的含量范围分别为0.825~1.7、5.15~10.05、0.34~1.95、0~0.002、0.198~2.62 mg·g^(-1)。结论:该方法操作简单,快速准确,灵敏度高,重复性好,可用于藤茶中5个黄酮类成分的含量测定。 Objective:To establish an HPLC-MS/MS method for simultaneous determination of five flavonoids in leaves of Ampelopsis grossedentata.Methods:The separation was performed on a CNW Athena C18-WP(100 mm×2.1 mm,3μm)column with gradient elution of methanol-0.1%formic acid aqueous solution at a flow rate of 0.3 mL·min^(-1)and a column temperature of 30℃.Negative ion detection was adopted with electrospray ion source and multiple reaction monitoring mode.Results:The linear ranges of myricetin,taxifolin,quercetin 3-rhamnoside,kaempferol and(-)-epicatechin gallat were 54.4-1030 ng·mL^(-1),28.4-1000 ng·mL^(-1),27.4-1000 ng·mL^(-1),24.8-998 ng·mL^(-1)and 30.6-1010 ng·mL^(-1)(r>0.9968),respectively.The RSDs of precision,stability and repeatability tests were below 5.0%(n=6),the average recoveries were 96.4%,99.8%,97.7%,95.0%and 100.9%with RSDs of 4.7%,1.4%,1.1%,4.3%and 3.0%(n=6),respectively.The average contents of the above 5 components in 4 samples were 0.825-1.7,5.15-10.05,0.34-1.95,0-0.002 and 0.198-2.62 mg·g^(-1),respectively.Conclusion:The method is simple,rapid,accurate,sensitive and repeatable,which can be used for the determination of five flavonoids in leaves of Ampelopsis grossedentata.
