芪术颗粒对肝纤维化模型大鼠肝脏细胞自噬的影响 被引量:1

Effects of Qizhu Granules on Autophagy in Hepatocytes of Rats with Liver Fibrosis

摘要 目的观察芪术颗粒对四氯化碳(carbon tetrachloride,CCl4)诱导大鼠肝纤维化模型自噬的影响。方法将40只Wistar大鼠随机分为正常对照组、模型对照组、复方鳖甲软肝片组、芪术颗粒组,除正常对照组外其余各组均腹腔注射CCl4建立肝纤维化模型,按照组别对应处理。4周后取肝组织,Western blot法检测α-平滑肌肌动蛋白(α-smooth muscle actin,α-SMA)及自噬相关基因3(autophagy-related gene 3,Atg3)、自噬相关基因Beclin1介导的蛋白表达,Real-time PCR检测Atg3、Beclin1基因表达,电镜下观察自噬情况。结果与正常对照组比较,模型对照组大鼠肝α-SMA蛋白显著增多(P<0.01);与模型对照组比较,复方鳖甲软肝片组、芪术颗粒组肝α-SMA蛋白显著减少(P<0.05);与正常对照组比较,模型对照组大鼠肝Atg3蛋白表达显著增加(P<0.01),Atg3基因表达增加,Beclin1蛋白及基因表达增加,自噬溶酶体和自噬体数量增多;与模型对照组比较,芪术颗粒组、复方鳖甲软肝片组Atg3蛋白及基因表达降低(P<0.05),芪术颗粒组Beclin1蛋白及基因表达降低(P<0.05),芪术颗粒组、复方鳖甲软肝片组组织中自噬溶酶体、自噬体数量减少。结论芪术颗粒可通过抑制肝脏细胞Atg3、Beclin1表达而影响自噬从而发挥其抗肝纤维化作用。 Objective To observe the effect of Qizhu Granules on autophagy in a rat liver fibrosis model induced by carbon tetrachloride(CCl4).Methods Forty Wistar rats were randomly divided into the normal control group,the model control group,the compound Biejia Ruangan Tablet group and the Qizhu Granule group.Except the normal control group,all other groups were intraperitoneally injected with CCl4 to establish liver fibrosis models,which were treated according to the corresponding groups.Liver tissue was taken after 4 weeks and detected by the method of western blotα-Smooth muscle actin(α-smooth muscle actin,α-SMA),autophagy related gene 3(Atg3),and autophagy related gene Beclin1(Beclin1).The genes expression of Atg3 and Beclin1 were detected by q-PCR,and autophagy was observed under electron microscope.Results Compared with the normal control group,the levels ofα-smooth muscle actin,α-SMA protein in liver of rats in the model control group were increased significantly(P<0.01);compared with the model control group,the liver of the compound Biejia Ruangan tablet group and Qizhu granule groupα-smooth muscle actin,α-SMA protein was decreased significantly(P<0.05);compared with the normal control group,the expression of Atg3 protein in the liver of the model control group was significantly increased(P<0.01),the expression of Atg3 gene,the expression of Beclin1 protein and gene,and the number of autophagic lysosomes and autophagosomes were increased;compared with the model control group,the expression of Atg3 protein and gene in Qizhu Granule group and Compound Biejia Ruangan Tablet group were decreased(P<0.05),the expression of Beclin1 protein and gene in Qizhu Granule group were decreased(P<0.05),and the number of autophagic lysosomes and autophages in the Qizhu Granule group and the Compound Biejia Ruangan Tablet group were decreased.Conclusion Qizhu Granule may exert its anti-hepatic fibrosis effect by inhibiting liver cell autophagy.
