高迁移率族蛋白1对失血性休克大鼠缺血再灌注后肠屏障功能影响 被引量:2

Effects of high-mobility group box-1 on intestinal barrier function in hemorrhagic rats after reperfusion

摘要 目的 探究高迁移率族蛋白1(HMGB1)对失血性休克大鼠缺血再灌注后肠屏障功能的影响。方法 Wistar大鼠32只,随机分为常温复苏(N)组和低温复苏(H)组,每组各16只。放血20 ml/kg,构筑失血性休克模型,90 min后,N组采用加热设备控制大鼠体温在38℃,H组采用酒精擦浴等方式控制大鼠体温在32℃。复苏后3 h,每组各处死8只大鼠,采集血液、肠系膜淋巴结及小肠标本,剩余鼠用于72 h生存分析。采用定量聚合酶链反应及蛋白印迹分别检测两组大鼠肠黏膜中HMGB1 mRNA及蛋白的表达。采用酶联免疫吸附试剂盒检测大鼠血清炎性因子表达情况,且进行小肠细菌移位检测,分析复苏后大鼠肠屏障功能改变情况。结果 H组大鼠小肠组织中HMGB1 mRNA表达水平明显低于N组,差异有统计学意义(P<0.05)。H组大鼠小肠组织中HMGB1、白细胞介素(IL)-4、神经丝蛋白-M、肾连蛋白、酰基辅酶A氧化酶2、IL-17、胰岛素样生长因子-1受体、淀粉样蛋白前体蛋白结合家族A2、肌醇需求酶1α、Bax、CLP和白细胞共同抗原蛋白表达明显低于N组,而Coronin 1A和IL-10蛋白表达明显高于N组,差异均有统计学意义(P<0.05)。H组大鼠血清中HMGB1、肿瘤坏死因子-α、IL-1β、IL-6和IL-8明显低于N组,抑炎因子IL-10高于N组,差异有统计学意义(P<0.05)。H组大鼠肠系膜淋巴结平均细菌移位量低于N组,差异有统计学意义(P<0.05)。72 h生存分析结果显示,N组大鼠平均生存时间为(54.6±4.8)h,低于H组的(75.79±3.62)h,差异有统计学意义(P<0.05)。结论 HMGB1的降低可能是诱导性低温复苏减轻大鼠失血性休克肠屏障功能损伤的重要分子机制。 Objective To investigate the effects of high-mobility group box-1(HMGB1)on intestinal barrier function after ischemia reperfusion in hemorrhagic shock rats.Methods Thirty-two Wistar rats were randomly divided into normal resuscitation group(N)and low resuscitation group(H),with 16 rats in each group.Hemorrhagic shock model was established by exsanguination of 20 ml/kg.Ninety minutes later, the body temperature of rats in group N was controlled at 38℃ by heating equipment, and the body temperature of rats in group H was controlled at 32℃ by alcohol bath.At 3 hours after resuscitation, 8 rats in each group were sacrificed, blood, mesenteric lymph nodes and small intestine samples were collected, and the remaining rats were used for 72 hours survival analysis.The expression of HMGB1 mRNA and protein in intestinal mucosa of the two groups were detected by quantitative polymerase chain reaction and western blot.Enzyme-linked immuno sorbent assay kit was used to detect the expression of inflammatory factors in serum of rats, and intestinal bacterial translocation detection was performed to analyze the changes of intestinal barrier function after resuscitation.Results The mRNA expression level of HMGB1 in small intestine of group H was significantly lower than that of group N,with statistical significance(P<0.05).The expressions of HMGB1,interleukin(IL)-4,Neurofilament-M,Nephronection, palmitoyl CoA oxidase 2(ACOX2),IL-17,insulin-like growth factor-1 receptor(IGF1R),the amyloid precursor protein-binding protein A2 gene(APBA2),inositol-requiring enzyme 1α(IRE1α),Bax, CLP and CD45 in small intestine of group H were significantly lower than those in group N,and Coronin 1A and IL-10 were significantly higher than those in N group, the difference was statistically significant(P<0.05).The serum levels of HMGB1,tumor necrosis factor-α(TNF-α),IL-1β,IL-6 and IL-8 in group H were significantly lower than those in group N,and the anti-inflammatory factor IL-10 was higher than that in group N,with statistical significance(
