白术药材UPLC特征图谱方法构建及模式识别研究 被引量:1

摘要 目的:以有机酸类物质为指标建立白术药材的超高效液相色谱指纹图谱测定方法,并结合聚类分析及主成分分析法(PCA)评价不同产地的白术药材质量。方法:以十八烷基硅烷键合硅胶为填充剂(柱长为50 mm,内径为2.1 mm,粒径为1.7μm),以乙腈为流动相A,以0.1%磷酸溶液为流动相B,进行梯度洗脱,流速为0.25 mL/min;检测波长为325 nm。指纹图谱采用中药色谱指纹图谱相似度评价(2012版)软件,对不同产地的白术UPLC指纹图谱进行分析,并运用SPSS软件对指纹图谱进行统计分析。结果:指纹图谱共标定了9个峰作为共有峰,并指认其中7个共有峰,分别为绿原酸、新绿原酸、隐绿原酸、1,3-二咖啡酰奎宁酸、异绿原酸B、异绿原酸A和异绿原酸C,15批白术药材相似度均大于0.8。聚类分析将15批白术分为5类,PCA结果表明,前2个主成分的累积贡献率达到81.72%,选择这2个主成分因子即可对白术药材进行综合评价。根据综合得分可知,来自安徽亳州和河北安国产地的白术药材中有机酸类成分占比更大,其质量可能更佳。结论:以有机酸类物质为指标建立的UPLC方法简单、可靠,精密度、稳定性、重复性良好,结合PCA可以快速、客观地评价不同产地白术药材的质量差异,为白术水提制剂的质量评价研究奠定基础。 Objective:An ultra-high performance liquid chromatography(UPLC) fingerprint determination method of Atractylodes macrocephala was established with organic acids as indicators, and the quality of Atractylodes macrocephala was evaluated by combining cluster analysis and principal component analysis(PCA).Methods:Octadecylsilane-bonded silica gel was uesd as a filler(50 mm×2.1mm, 1.7μm),acetonitrile was used as mobile phase A,and 0.1% phosphoric acid solution was used as mobile phase B to perform gradient for elution, the flow rate was 0.25ml per minute;the detection wavelength was 325 nm.The UPLC fingerprints of Atractylodes macrocephala from different origins were analyzed by using the similarity evaluation software of Chinese medicine chromatographic fingerprints(Version 2012),and the SPSS software was applied to perform statistical analysis on fingerprints.Results:A total of 9 peaks were calibrated as common peaks in the fingerprint spectrum, and 7 of them were identified as chlorogenic acid, neochlorogenic acid, cryptochlorogenic acid, 1,3-dicaffeoylquinic acid, isochlorogenic acid B,isochlorogenic acid A and isochlorogenic acid C.The similar degrees of 15 batches of Atractylodes macrocephala is greater than 0.80.Cluster analysis classified 15 batches of Atractylodes macrocephala into five categories, and the result of PCA showed that the accumulative contribution rate of the first two principal components was 81.72%,so as long as the two factors were chosen, the quality of Atractylodes macrocephala could be evaluate basically.According to the composite score, the quality of Atractylodes macrocephala from the areas of Anhui Bozhou and Hebei Anguo account for a larger proportion of organic acids, and their quality may be better.Conclusion:The established method using organic acids as indicators is simple and reliable, with good precision, stability and repeatability.The application of UPLC combined with PCA could quickly and objectively evaluate the quality of Atractylodes macrocephala form different regions, a
