摘要 目的 优化HD-BioP3细胞的电转染条件,提高HD-BioP3细胞的电转染效率。方法 使用电转染的方法将GFP质粒转入HD-BioP3细胞内,对电转染缓冲液、电转染温度和电转染质粒量3个因素进行优化,24 h后通过全自动荧光细胞分析仪检测细胞活率及电转染效率。结果 3个因素中,电转染缓冲液对电转染效率影响最大,其次为电转染质粒量,电转染温度影响最小。在EX-CELL?CD CHO Fusion作为电转染缓冲液、电转染温度为4℃、电转染质粒量为30μg的条件下,电转染效果最好。结论 优化HD-BioP3细胞的电转染条件,可为研究外源基因电转染HD-BioP3细胞提供基础数据。 Objective The electrotransfection conditions of HD-BioP3 cells were optimized to improve the electrotransfection efficiency. Methods With the transfer of GFP plasmid into HD-BioP3 cells by electrotransfection,the three factors(electrotransfection buffer,electrotransfection temperature and quantity of electrotransfection plasmid) were optimized. After 24 hours, the cell viability and electrotransfection efficiency were detected by automatic fluorescence cell analyzer. Results Among the three factors, electroporation buffer had the greatest effect on transfection efficiency, followed by plasmid quantityn, and electroporation temperature had less effect. With EX-CELL?CD CHO Fusion as electrotransfection buffer, electrotransfection temperature at 4 ℃ and electrotransfection plasmid quantity 30 μg, the effect of electrotransfection was the best.Conclusion With the optimization of electrotransfection conditions of HD-BioP3 cells, basic data can be provided in the study of foreign gene electrotransfection of HD-BioP3 cells.
