摘要
目的探讨结直肠癌患者血清长链非编码RNA(lncRNA)小核RNA宿主基因11(SNHG11)及微小RNA(miRNA)miR-27b的表达及与预后的关系。方法将2016年1月至2018年1月广州市增城区中医医院收治的94例结直肠癌患者纳入研究作为结直肠癌组,将同期于该院就诊的94例结直肠良性疾病患者纳入研究作为结直肠癌良性疾病组,另选取94例体检健康者作为对照组。比较3组血清lncRNA SNHG11、miR-27b水平。结直肠癌组患者根据临床特征分为不同的亚组,比较亚组间lncRNA SNHG11、miR-27b水平。随访3年,根据患者存活情况分为存活组和死亡组,比较两组lncRNA SNHG11、miR-27b水平。采用受试者工作特征(ROC)曲线分析lncRNA SNHG11、miR-27b水平用于评估患者预后的价值。以结直肠癌患者lncRNA SNHG11、miR-27b水平平均值为界将患者分为高、低表达组,采用K-M生存曲线分析两组患者累积存活率。结果相较于对照组及结直肠良性疾病组,结直肠癌组患者血清lncRNA SNHG11水平更低,而miR-27b水平更高(P<0.05);亚组分析结果显示:患者lncRNA SNHG11、miR-27b水平与淋巴结转移及TNM分期有关(P<0.05)。相较于存活组,死亡组lncRNA SNHG11水平更低,而miR-27b水平更高(P<0.05)。ROC曲线分析显示:lncRNA SNHG11用于评估患者生存预后的最佳截断值为3.883,曲线下面积为0.868;miR-27b用于评估患者生存预后的最佳截断值为3.971,AUC为0.851。随访3年,lncRNA SNHG11高表达组(54例)中36例存活,累积存活率为66.67%,lncRNA SNHG11低表达组(40例)中16例存活,累积存活率为40.00%,两组累积存活率比较差异有统计学意义(P<0.05);miR-27b高表达组(43例)中18例存活,累积存活率41.86%,miR-27b低表达组(51例)中34例存活,累积存活率为66.67%,两组累积存活率比较差异有统计学意义(P<0.05)。结论结直肠癌患者血清lncRNA SNHG11水平下降,而miR-27b水平升高,二者水平与患者淋巴结转移、TNM分期有关,可作为患者生存�
Objective To investigate the expression of serum long non-coding RNA(lncRNA)small nuclear RNA host gene 11(SNHG11)and microRNA(miRNA)miR-27b in patients with colorectal cancer and their relationship with prognosis.Methods A total of 94 patients with colorectal cancer admitted to Guangzhou Zengcheng Hospital of Traditional Chinese Medicine from January 2016 to January 2018 were enrolled in the study as the colorectal cancer group.Ninety-four patients with benign colorectal diseases who were treated in the hospital during the same period were enrolled in the study as the colorectal cancer benign disease group.In addition,94 healthy people were enrolled as the control group.The serum levels of lncRNA SNHG11 and miR-27b were compared among the three groups.Patients in the colorectal cancer group were divided into different subgroups according to clinical characteristics,and the levels of lncRNA SNHG11 and miR-27b were compared between subgroups.The patients were followed up for 3 years,and they were divided into survival group and death group according to their state of survival,and the levels of lncRNA SNHG11 and miR-27b were compared between the two groups.The receiver operating characteristic(ROC)curve was used to analyze the value of lncRNA SNHG11 and miR-27b levels in evaluating the prognosis of patients.Based on the average levels of lncRNA SNHG11 and miR-27b in colorectal cancer patients,the patients were divided into high-and low-expression groups,and the cumulative survival rate of the two groups was analyzed by K-M survival curve.Results Compared with the control group and the colorectal benign disease group,the serum level of lncRNA SNHG11 in the colorectal cancer group was lower,while the level of miR-27b was higher(P<0.05).Subgroup analysis showed that the levels of lncRNA SNHG11 and miR-27b were related to lymph node metastasis and TNM stage(P<0.05).Compared with the survival group,the death group had significantly lower level of lncRNA SNHG11 and a significantly higher level of miR-27b(P<0.05).ROC curv
