摘要
目的:探讨二甲双胍对类风湿关节炎(RA)成纤维样滑膜细胞(FLS)增殖及分泌炎性细胞因子的影响。方法:取我院行关节镜手术治疗的RA患者关节滑膜组织,采用改良组织块法培养原代FLS,用不同浓度二甲双胍(10、20、30μmol/L)干预FLS细胞,分别记为小剂量、中剂量、高剂量组,以常规养的FLS细胞为对照组。采用CCK-8法检测各组细胞处理24h、48h、72h后细胞增殖情况,采用ELISA检测各组细胞处理72h后上清液中促炎细胞因子:肿瘤坏死因子-α(TNF-α)、白细胞介素-17(IL-17)、白细胞介素-6(IL-6)和抗炎细胞因子:白细胞介素-10(IL-10)水平,比较组间各指标水平的差异。结果:与对照组相比,72h后,中剂量组、高剂量组FLS增殖水平均降低(P<0.05);高剂量组上清液中TNF-α、IL-6、IL-17水平降低,IL-10水平升高(P<0.05)。结论:高剂量二甲双胍具有抑制FLS增殖和促炎细胞因子分泌、促进抗炎细胞因子分泌的作用,对治疗RA具有一定的应用价值。
Objective:To investigate the effect of metformin on the proliferation and secretion of inflammatory cytokines of fibroblast-like synovial cells(FLS)in rheumatoid arthritis(RA).Method:The synovial tissue of RA patients treated by arthroscopic surgery in our hospital was taken,and the primary FLS was cultured with modified tissue block method,and different concentrations of metformin(10,20,30μmol/L)to interfere with FLS cells,which were recorded as low-dose,medium-dose and high-dose groups respectively,and the conventionally raised FLS cells were used as the control group.CCK-8 method was used to detect the proliferation of cells in each group after treatment for 24h,48h and 72h.The levels of pro-inflammatory cytokine tumor necrosis factor-α(TNF-α),interleukin-17(IL-17),interleukin-6(IL-6)and anti-inflammatory cytokine interleukin-10(IL-10)in the supernatant of each group after 72h treatment were detected by ELISA,and the levels of each index were compared between groups.Results:Compared with the control group,the proliferation level of FLS in the middle dose group and the high dose group were decreased after 72hours(P<0.05);The levels of TNF-α,IL-6and IL-17in supernatant of high-dose group were decreased and the level of IL-10was increased(P<0.05).Conclusion:High-dose metformin can inhibit the proliferation of FLS,promote the secretion of inflammatory cytokines,and promote the secretion of anti-inflammatory cytokines.It has certain application value in the treatment of RA.
