摘要
试验旨在探究厚朴提取物对产气荚膜梭菌感染肉鸡生长性能、肠道组织形态和屏障功能的影响。将264只1日龄肉仔鸡随机分成4组,每组6个重复,每个重复11只。试验期为23 d。对照组和产气荚膜梭菌感染组饲喂基础饲粮,厚朴提取物组和厚朴提取物+产气荚膜梭菌感染组在基础饲粮中添加400 mg/kg厚朴提取物。于18~23日龄产气荚膜梭菌感染组和厚朴提取物+产气荚膜梭菌感染组口服接种A型产气荚膜梭菌悬浮液1 mL(1×10^(8)cfu/mL,每天1次),对照组和厚朴提取物组肉鸡灌服等量的灭菌培养基。结果显示:产气荚膜梭菌感染造成肉鸡小肠黏膜损伤,提高了23日龄回肠损伤评分(P<0.05)和血清二胺氧化酶活力,并降低了十二指肠绒隐比(P<0.05)及肉鸡空肠sIgA含量(P<0.05);而日粮中添加厚朴提取物有降低1~17日龄肉鸡平均日采食量的趋势(P=0.05),降低了23日龄肉鸡空肠绒毛高度(P<0.05),提高了肉鸡空肠sIgA含量(P<0.05);产气荚膜梭菌感染下调了空肠occludin、claudin-2及回肠mucin5ac、occludin、claudin-2 mRNA表达量(P<0.05),而添加厚朴提取物可提高产气荚膜梭菌感染肉鸡回肠mucin5ac mRNA表达量(P<0.05)。因此,饲粮中添加厚朴提取物可改善产气荚膜梭菌感染肉鸡肠道形态结构和免疫功能,调控肠道屏障功能相关基因表达。
The current study was conducted to investigate the effects of Magnolia officinalis extract on the growth performance,intestinal morphology and barrier function in broilers challenged with Clostridium perfringens(C.perfringens).A total of2641-day-old broilers were randomly divided to 4 treatment groups with 6 replicates in each group and 11 birds in each replicate.The trial period lasted for 23 days.Broilers in the control group and C.perfringens infection group were fed with basal diet,while chicks in the Magnolia officinalis extract and Magnolia officinalis extract+C.perfringens infection groups were fed with basal diet supplemented with 400 mg/kg of Magnolia officinalis extract.From 18 to 23 days of age,broilers in the C.perfringens infected and Magnolia officinalis extract+C.perfringens infection groups were continuously oral inoculated with 1 mL suspension of type A C.perfringens(1×10^(8)cfu/mL,once a day),whereas those in the control and Magnolia officinalis extract groups were treated with the same volume of sterile medium.The results showed that C.perfringens infection caused damage to the small intestinal mucosa in birds,and increased(P<0.05)the ileal injury lesion score at the 23 days of age,as well as the activity of serum diamine oxidase,but decreased the ratio of duodenal villus height/crypt depth and jejunal secretory immunoglobulin A(sIgA)level(P<0.05).However,supplementation of Magnolia officinalis extract in diet tended to decreased the average daily feed intake(ADFI)of broilers from 1 to 17 days of age(P=0.05).Moreover,Magnolia officinalis extract decreased the jejunal crypt depth(P<0.05),but increased the sIgA concentration in the jejunum of broilers at the 23 days of age(P<0.05).The qPCR results further demonstrated that C.perfringens infection down-regulated the mRNA expression for occludin and claudin-2 in the jejunum(P<0.05),as well as mucin5ac,occludin,and claudin-2 in the ileum of birds.Mucin5ac mRNA level in the ileum was elevated in the C.perfringens-infected broilers fed with Magnolia
