基于p38 MAPK信号通路探讨穴位埋线对哮喘大鼠肺组织Th1/Th2失衡及EOS的影响 被引量:3

摘要 目的观察穴位埋线对哮喘大鼠肺组织磷酸化p38丝裂原活化蛋白激酶(p-p38 mitogen-activated protein kinase,p-p38 MAPK)、γ-干扰素(interferon-γ,IFN-γ)、白细胞介素-4(interleukin-4,IL-4)的表达和嗜酸性粒细胞(eosinophil,EOS)的影响,探讨穴位埋线治疗哮喘的作用机制。方法将40只Wistar雄性大鼠随机分为正常组、模型组、西药组和穴位埋线组,采用卵蛋白和氢氧化铝混悬液制备哮喘模型,西药组连续2周给予地塞米松腹腔注射治疗,穴位埋线组于肺俞、肾俞、脾俞、定喘和膻中处行埋线治疗,观察各组大鼠引喘潜伏期的变化,HE染色观察肺组织病理形态学的变化,透射电镜观察肺组织中EOS超微结构的变化,瑞氏染色观察肺泡灌洗液(bronchoalveolar lavage fluid,BALF)中EOS的相对含量,Western blot检测肺组织中p-p38 MAPK、IFN-γ、IL-4蛋白的表达。结果模型组大鼠支气管形态结构异常,大鼠肺组织EOS处于活化状态,西药组和穴位埋线组支气管变形程度减轻,大鼠肺组织EOS出现细胞自噬样结构的变化。与正常组比较,模型组大鼠引喘潜伏期明显缩短(P<0.01),大鼠BALF中EOS的相对含量、肺组织中p-p38 MAPK和IL-4蛋白的表达量明显升高(P<0.01),而IFN-γ蛋白的表达量明显降低(P<0.01);与模型组比较,西药组和穴位埋线组大鼠的引喘潜伏期明显延长(P<0.01),大鼠BALF中EOS的相对含量、肺组织中p-p38 MAPK和IL-4蛋白的表达量明显降低(P<0.01),而IFN-γ蛋白的表达量明显升高(P<0.01)。结论穴位埋线可以通过抑制p38 MAPK信号通路的传导,纠正Th1/Th2失衡,从而影响肺组织中EOS的浸润和活化,发挥治疗哮喘的作用。 Objective To study the effects of catgut embedding at acupoints on the expression of phosphorylated p38 MAPK,IFN-γ,IL-4 and EOS in the lung tissue of asthmatic rats,and explore the mechanism of catgut embedding at acupoints in the treatment of asthma.Methods 40 Wistar male rats were randomly divided into normal group,model group,western medicine group and acupoint embedding group.The asthma model was prepared with ovalbumin and aluminum hydroxide suspension.The western medicine group was given intraperitoneal injection of dexamethasone for 2 consecutive weeks,and the acupoint embedding group received embedding at Feishu,Shenshu,Pishu,Dingchuan and Danzhong.The incubation period of asthma in each group was observed,HE staining was used to observe the pathological changes of lung tissue,transmission electron microscope was used to observe the changes of EOS ultrastructure in lung tissue,Wright-Giemsa staining was used to observe the relative content of EOS in bronchoalveolar lavage fluid(BALF),Western blot was used to detect the expression of p-p38 MAPK,IFN-γand IL-4 protein in lung tissue.Results The morphology and structure of the bronchus in the model group were abnormal,and the EOS in the lung tissue of the rats was in an activated state;the degree of bronchial deformation was reduced,and the EOS in the lung tissue showed the change of the autophagy bubble-like structurein the western medicine group and the acupoint embedding group.Compared with the normal group,the incubation period of asthma in the model group was significantly shortened(P<0.01),and the relative content of EOS in rat BALF,the expression of p-p38 MAPK and IL-4 protein in lung tissue were significantly increased(P<0.01),while the expression of IFN-γprotein was significantly reduced(P<0.01).Compared with the model group,the incubation period of asthma was significantly prolonged(P<0.01),the relative content of EOS in the rat BALF,the expression of p-p38 MAPK and IL-4 protein in the lung tissue were significantly reduced(P<0.01),while the expr
