西门塔尔牛无角性状的分子鉴定及应用

摘要 为鉴定西门塔尔牛有角和无角性状的相应基因型,为西门塔尔牛无角品系的培育提供数据基础,试验采集了103头无角西门塔尔牛、5头有角西门塔尔牛和5头无角安格斯牛颈静脉血,提取全血DNA后,通过PCR-测序法检测牛PC基因中P202ID突变位点的基因型。分型与测序结果显示,108头西门塔尔牛P202ID位点的基因型与角型完全一致,其中,103头无角西门塔尔牛中4头为纯合子,基因型与无角安格斯牛一致,均为PC/PC,PC的基因频率为0.0370;82头为无角杂合子,基因型为PC/prs,其中,PC的基因频率为0.7593;剩余17头去角西门塔尔牛与5头有角西门塔尔牛基因型一致,均为prs/prs,prs的基因频率为0.2037。鉴定群体中PC的基因频率为0.4166,prs的基因频率为0.5834。卡方检验结果显示,P202ID基因型与检测群体中有无角性状具有极显著关联,可以利用P202ID位点对西门塔尔牛角型进行鉴定。 In order to identify the corresponding genotypes of the horned and hornless traits of Simmental cattle,then provide data basis for breeding hornless line of Simmental cattle,in this study,venous blood of 103 hornless Simmental cattle,five horned Simmental cattle,and five hornless Angus cattle was collected,with the whole blood DNA extracted,then the P202IDmutation site of bovine PCgene was genotyped by PCR-sequencing.The results showed that the phenotypes of 108 Simmental cattle were completely consistent with their genotypes of P202IDlocus,among 103 hornless Simmental cattle,four were homozygous hornless cattle,whose genotype was consistent with that of hornless Angus cattle,it was PC/PC,and the genotypic frequency of PCwas 0.0370;82 were heterozygous hornless Simmental cattle,whose genotype was PC/prs,and the genotypic frequency of PCwas 0.7593;the genotype of 17 mulled Simmental and 5 horned Simmental were all prs/prs,and the genotypic frequency was 0.2037.Of all the cattle used for genotyping,the gene frequency of PCwas 0.4166,and the frequency of prs was0.5834.Chi-square test showed that the genotype of P202IDlocus was significantly associated with horned traits(P<0.001),and P202IDlocus could be used to identify Simmental horn trait.
