基于文献的中药治疗肝癌实验研究现状分析 被引量:5

Experimental Research on Liver Cancer Treated by Traditional Chinese Medicine:A Literature Review

摘要 目的 梳理并探讨中药治疗肝癌实验研究的现状及未来研究方向。方法 计算机检索Cochrane Library、Web of Science、PubMed、中国期刊全文数据库、中文科技期刊全文数据库、中国生物医学文献数据库、万方医药期刊全文及学位论文数据库中的中药治疗肝癌实验研究,检索截止时间为2022年5月31日。提取纳入文献的发表时间、研究药物、肝癌模型、应用系统生物学技术等信息构建数据库并对提取信息进行统计分析。结果 共纳入文献735篇,包括中文文献612篇,英文文献123篇;涉及中药复方的研究532篇,药对18篇,单药185篇。使用频率排在前三位的复方依次是扶正抑瘤方(3.13%,23/735)、鳖甲煎丸(2.45%,18/735)、小柴胡汤(1.22%,9/735),排在前三位的药对分别是白花蛇舌草-半枝莲(0.41%,3/735)、当归-红芪(0.27%,2/735)、姜黄-海带(0.27%,2/735),排在前三位的单药依次是白花蛇舌草(2.31%,17/735)、半枝莲(1.22%,9/735)、丹参(0.95%,7/735)。肝癌模型为细胞株的文献328篇,动物模型的文献404篇;使用频率排在前三位的肝癌细胞株分别是HepG2 (21.63%,159/735)、SMMC-2771(10.61%,78/735)、H22 (3.67%,27/735),排在前三位的肝癌动物模型分别是皮下移植瘤小鼠(42.45%,312/735)、化学诱导肝癌模型(7.07%,52/735)、原位肝癌模型(2.31%,17/735)。有199篇文献采用了网络药理学技术,6篇文献应用转录组学,3篇应用基因芯片,3篇应用代谢组学,2篇应用肠道菌群16S rDNA测序,1篇应用蛋白质组学分析。结论 目前中药治疗肝癌实验研究存在研究药物数量较多但研究欠缺系统性、新兴生物学技术应用不充分等不足;今后研究应以整体性和系统性为指导,以明确药效物质基础为质控,充分运用系统生物学技术,以期取得突破性进展。 Objective To summarize the current situation of experimental research on liver cancer treated by traditional Chinese medicine(TCM),and discuss the future direction of research.Methods Databases including Cochrane Library,Web of Science,PubMed,CNKI,VIP,SinoMed,and Wanfang were searched from the inception date till May 31st,2022 for experimental research on liver cancer treated by TCM.The data of the included research,involving the publication time,drugs,liver cancer models,applied systems biology technology and others,were extracted to construct a database and taken for statistical analysis.Results A total of 735 papers were included,612 Chinese and 123 English papers.TCM compound formulas in 532 papers,herbal pairs in 18 papers and single herbs in 185 papers were explored.The top three compound formulas were Fuzheng Yiliu Formula(扶正抑瘤方,3.13%,23/735),Biejiajian Pill(鳖甲煎丸,2.45%,18/735),and Xiaochaihu Decoction(小柴胡汤,1.22%,9/735).The three most frequently explored herbal pairs were Herba Hedyotis Diffusae(Baihuasheshecao)-Herba Scutellariae Barbatae(Banzhilian)(0.41%,3/735),Radix Angelicae Sinensis(Danggui)-Radix Hedysari(Hongqi)(0.27%,2/735),and Rhizoma Curcumae Longae(Jianghuang)-Herba Zosterae Marinae(Haidai)(0.27%,2/735).The top three single herbs studied were Herba Hedyotis Diffusae(2.31%,17/735),Herba Scutellariae Barbatae(1.22%,9/735),and Radix et Rhizoma Salviae Miltiorrhizae(Danshen)(0.95%,7/735).In terms of liver cancer models used in the research,there were 328 papers on cell lines and 404 papers on animal models.The three most frequently used liver cancer cell lines were HepG2(21.63%,159/735),SMMC-2771(10.61%,78/735),and H22(3.67%,27/735),while the top three animal models were mice with subcutaneous transplanted tumors(42.45%,312/735),chemically induced liver cancer models(7.07%,52/735),and orthotopic liver cancer model(2.31%,17/735).There were 199 papers using network pharmacology technology,6 papers utilizing transcriptomics,3 papers adopting gene chips,3 papers applying metabo
