基于HPLC和网络药理学分析经典名方辛夷清肺饮的质量标准研究 被引量:4

摘要 目的:基于HPLC法和网络药理学建立辛夷清肺饮(XQY)的质量标准。方法:采用HPLC建立XQY的指纹图谱,同时测定栀子苷、芒果苷、异阿魏酸、黄芩苷、汉黄芩苷、黄芩素、甘草酸7个成分的含量;继以网络药理学筛选和分析7个定量成分治疗变应性鼻炎的作用靶点和通路,构建“成分-疾病-靶点-通路”网络,预测XQY发挥治疗变异性鼻炎(AR)的质量标志物(Q-Marker)及其作用机制。结果:15批XQY的HPLC指纹图谱相似度均>0.99,栀子苷、芒果苷、异阿魏酸、黄芩苷、汉黄芩苷、黄芩素、甘草酸的质量分数分别为138.69~224.58μg·ml^(-1)、11.28~21.20μg·ml^(-1)、3.26~5.62μg·ml^(-1)、338.55~725.02μg·ml^(-1)、48.59~90.27μg·ml^(-1)、2.18~4.13μg·ml^(-1)、9.26~18.23μg·ml^(-1)。经网络药理学分析,7个定量成分可预选为XQY治疗AR的Q-Marker,其治疗AR的作用靶点共有115个,主要富集在细胞对生长因子刺激的反应、蛋白激酶结合、炎症反应等生物过程,以及核因子κB(NF-κB)信号通路、环磷鸟苷-蛋白激酶G(cGMP-PKG)信号通路、环磷酸腺苷(cAMP)信号通路等。结论:本研究所建立的整体质量控制方法和预选的Q-Marker具有代表性,可用于评价XQY的质量,同时可为其他经典名方以及中药复方的质量标准研究提供示范。 Objective:To establish the quality standard for Xinyi Qingfei Yin(XQY)based on HPLC and network pharmacology.Methods:The fingerprint of XQY was established by HPLC.Furthermore,the contents of geniposide,mangiferin,isoferulic acid,baicalin,wogonoside,baicalein and glycyrrhizic acid were determined.After screening and analyzing the function targets and pathways of seven quantitative compounds for allergic rhinitis by network pharmacology,the compound-disease-target-pathway network was constructed to predict the Q-marker and mechanism of XQY in therapeutic effect on allergic rhinitis.Results:The similarity of HPLC fingerprints of 15 batches of XQY was more than 0.99.The mass fractions of geniposide,mangiferin,isoferulic acid,baicalin,wogonoside,baicalein and glycyrrhizic acid in 15 batches of XQY were within the ranges of 138.69-224.58μg·ml^(-1),11.28-21.20μg·ml^(-1),3.26-5.62μg·ml^(-1),338.55-725.02μg·ml^(-1),48.59-90.27μg·ml^(-1),2.18-4.13μg·ml^(-1)and 9.26-18.23μg·ml^(-1),respectively.According to the analysis of network pharmacology,seven quantitative compounds were selected as the potential Q-Marker in XQY.There were 115 targets of XQY in treating AR,and they were mainly concentrated in biological processes such as cellular response to growth factor stimulus,protein kinase binding,inflammatory response,and NF-κB signaling pathway,cGMP-PKG signaling pathway and cAMP signaling pathway,etc.Conclusion:The whole quality control method and the preselected Q-marker in this study are representative,which can be used to evaluate the quality of XQY.Meanwhile,it also can serve as an example for the study of quality standards for other classical prescriptions and traditional Chinese medicine compound prescriptions.
