摘要
白桦(Betula platyphylla)是一种分布广泛的落叶乔木,具有重要的生态态、药用、经济价值。白桦在幼苗期常受到各种病原菌侵害,因此其抗病育种成为一个重要课题。类甜蛋白(thaumatin-like protein,TLP)是一类与植物抗病性相关的蛋白,本研究克隆了白桦类甜蛋白BpTLP35基因,对BpTLP35进行生物信息学分析,结果表明,白桦BpTLP35共有225个氨基酸,理论等电点为8.30,是一种亲水、稳定的蛋白质,具有信号肽结构,亚细胞定位在液泡中;BpTLP35基因的启动子含有茉莉酸甲酯、脱落酸等激素响应元件,还含有光、低温和机械损伤响应等相关元件,说明BpTLP35与白桦逆境胁迫密切相关;组织表达特异性分析结果表明,BpTLP35基因主要在花、木栓层、木质部和叶片中表达;在NaCl、低温和病原菌胁迫下,BpTLP35基因均上调表达,说明BpTLP35与白桦抗生物和非生物胁迫均密切相关;通过Gateway技术将该基因克隆到pGWB2植物表达载体上,为白桦BpTLP35基因功能的进一步研究提供参考。
Betula platyphylla is a type of widely distributed deciduous trees with great ecological,medicinal and economic values.B.platyphylla is often attacked by various pathogens in its seedling stage,making the resistance breeding an important project.Thaumatin-like proteins(TLPs)are a kind of proteins related to plant disease resistance.In this study,BpTLP35 was cloned.Bioinformatics analysis shows that Bp TLP35 contains 225 amino acids with a theoretical isoelectric point of 8.30.It is a hydrophilic,stable protein with signal peptide and subcellular location in vacuoles.The promoter of Bp TLP35 contains response elements to low temperature,light,abscisic acid,methyl jasmonate and mechanical hurt,indicating that BpTLP35 is closely related to the stress response in B.platyphylla.Tissue-specific expression analysis shows that Bp TLP35 mainly expressed in flowers,phellems,xylems and leaves.Bp TLP35 was up-regulated under NaCl stress,low temperature stress and pathogen infection,indicating that BpTLP35 is closely related to biotic and abiotic stress response.BpTLP35 is successfully cloned to pGWB2 vector through Gateway method.This study would provide reference to the studies on BpTLP35 functions in B.platyphylla.
