摘要
目的观察电针对功能性消化不良(FD)模型大鼠的治疗作用并筛查电针治疗前后差异表达的miRNA,进行生物信息学分析,以探索miRNA参与电针治疗FD过程中的相关分子机制。方法采用SPF级健康雄性SD大鼠18只,随机分为空白对照组、模型组和电针组,每组6只。所有造模大鼠均采用多因素应激干预法构建FD模型,造模成功后电针组采用电针足三里穴和太冲穴治疗。观察整个实验过程中大鼠的一般情况;在治疗结束后对各组大鼠的胃排空率进行检测;通过提取电针组和模型组大鼠总RNA,进行差异表达miRNA的筛选及生物信息学分析。结果在造模期间,造模大鼠出现饮食、饮水量和体重下降,毛发脱落变黄,大便稀溏,精神萎靡,活动量减少;电针治疗后电针组大鼠饮食、饮水、精神、活动度等情况好转,毛发变白,大便正常。模型组和电针组胃排空率明显低于空白对照组,而电针组胃排空率明显高于模型组,3组间比较,差异均具有统计学意义(F=185.691,P<0.01)。筛查出电针组和模型组大鼠显著差异表达的miRNA共16个,其中上调miRNA 7个和下调miRNA 9个。预测到差异表达的miRNA调控的靶基因数为11678个,靶位点数为31974个;KEGG通路富集分析最主要的20条信号通路以及GO功能富集分析排列最靠前的20个功能,其中生物过程的占比最高,为74.9%。结论电针能够有效改善FD模型大鼠的胃动力,并且此过程中存在多种miRNA参与其相关分子机制,该研究为FD的针灸治疗以及治疗靶点的选择提供了一定的理论依据。
Objective To study the therapeutic effect of electroacupuncture on functional dyspepsia(FD)rats,the differential expression of miRNA before and after electroacupuncture treatment was screened and bioinformatics analysis was conducted to explore the molecular mechanisms of miRNA involved in electroacupuncture treatment of FD.Methods Eighteen SPF healthy male SD rats were randomly divided into control group,model group and electroacupuncture group,with 6 rats in each group.FD rat model was constructed by multifactor stress intervention method in all model rats.After the model was successfully built,electroacupuncture group was treated with electroacupuncture ST36 and LR3.The general condition of rats was observed during the whole experiment.After treatment,gastric emptying rate of rats in each group was detected.Total RNA was extracted from the electroacupuncture group and the model group for screening and bioinformatics analysis of differentially expressed miRNA.Results During the modeling period,the model rats showed decreased diet,water intake and weight,hair loss and becoming yellow,loose stool,listlessness and decreased activity.After electroacupuncture treatment,food and water intake,mental activity and other conditions of the electroacupuncture group rats improved,hair turned white,and stool normal.Gastric emptying rate of the model group and electroacupuncture group was significantly lower than that of the control group,while gastric emptying rate of the electroacupuncture group was significantly higher than that of the model group,and the differences among the three groups were statistically significant(F=185.691,P<0.01).A total of sixteen miRNAs were significantly differentially expressed in the electroacupuncture group and the model group,including 7 up-regulated miRNAs and 9 down-regulated miRNAs.It was predicted that the number of target genes regulated by differentially expressed miRNAs was 11678,and the number of target sites was 31974.KEGG pathway enrichment analysis of the 20 most important signali
