疏肝健脾解毒方中毛蕊异黄酮葡萄糖苷的含量测定 被引量:4

摘要 目的:建立高效液相色谱(HPLC)法测定疏肝健脾解毒方中毛蕊异黄酮葡萄糖苷的含量。方法:采用HPLC法,以依利特Supersil ODS-C18柱(4.6 mm×250 mm,5μm)为色谱柱,以乙腈-0.2%甲酸溶液为流动相梯度洗脱(0~23 min,12%乙腈;23~65 min,12%~26%乙腈),流速为1.0 ml/min,柱温为30℃,检测波长为260 nm。结果:经方法学验证,毛蕊异黄酮葡萄糖苷在7.2~43.2μg(R2=0.9998)范围内线性关系良好,精密度、稳定性和重复性试验的RSD均<2.0%(n=6),加样回收率为98.87%(RSD为1.79%)。结论:本研究建立的含量测定方法简便,结果准确可靠,适用于疏肝健脾解毒方中毛蕊异黄酮葡萄糖苷的测定,可为其质量标准的深入研究提供依据。 Objective:To establish a high-performance liquid chromatography(HPLC)method for determining the content of calycosin glucoside in liver-soothing,spleen-strengthening,and detoxicating prescription.Methods:HPLC was performed on an Elite Supersil ODS-C18 column(4.6 mm×250 mm,5μm)with a mobile phase of acetonitrile-0.2%formic acid solution for gradient elution(0-23 minutes:12%acetonitrile;23-65 minutes:12%-26%acetonitrile),at a flow rate of 1.0 ml/min,a column temperature of 30℃,and a detection wavelength of 260 nm.Results:Methodological validation showed that calycosin glucoside had a good linear relationship within the range of 7.2~43.2μg(R2=0.9998),and precision,stability,and reproducibility tests had a relative standard deviation(RSD)of<2.0%(n=6),with a recovery rate of 98.87%(RSD=1.79%).Conclusion:The method for content determination established in this study is simple and convenient with accurate and reliable results and is thus suitable for determining the content of calycosin glucoside in liversoothing,spleen-strengthening,and detoxicating prescription,which provides a basis for in-depth research on quality standard.
