云南不同地区大绒鼠种群的表型性状差异与分子遗传差异的比较与分析

摘要 为探究云南不同地区大绒鼠Eothenomys miletus的表型性状差异(PST)和分子遗传差异(FST),本研究采集了云南省5个不同地区(德钦、香格里拉、丽江、剑川和哀牢山)的73只大绒鼠,首先对14个表型数量性状进行差异分析,基于性状数据计算PST,并通过简化基因组学所获得的高质量SNPs,计算两两种群间的成对FST。结果发现,除下齿列长外,种群间的表型性状均存在显著差异;种群间的PST均大于FST;海拔差与种群间的体质量、体长、尾长、前肢长、最大眶间距和上齿列长等的PST呈正相关关系,冬季平均温度差与体质量、体长、尾长、前肢长和最大眶间距呈正相关关系,冬季总降水量差与躯干长、耳长的PST呈正相关关系,而地理距离与种群间各性状的PST无相关关系。这表明,云南不同地区大绒鼠的表型性状受到较明显的选择压力,而海拔、环境温度和降水量可能是影响其种群间表型分化的环境因子。 To explore the differentiation in phenotypic traits(PST)and differentiation in neutral marker(FST)among Eothenomys miletus individuals from different areas of Yunnan Province.In this study,73 E.miletus were collected from Deqin,Xianggelila,Lijiang,Jianchuan and Ailaoshan.We compared and analyzed PST using 14 phenotypic traits and high-quality SNP-based FST using specific-locus amplified fragment sequencing.The results showed that there were significant differences in phenotype traits among the 5 populations except the lower tooth row length,and most of the traits were PST>FST;a positive correlation between altitude difference and PST of body weight,body length,tail length,forelimb length,interorbital breadth and upper tooth length was detected among populations,while winter temperature difference was positively correlated with the PST of body weight,body length,tail length,forelimb length and interorbital breadth,and winter precipitation difference was also positively correlated with the PST of body length and ear length.This study suggests that directional selection occurred in E.miletus populations in Yunnan Province,and altitude,temperature and precipitation may be the environmental factors influencing the differentiation of phenotypic traits.
