摘要
目的:探究木犀草素(luteolin,LT)通过TLR4/NF-κB途径对OVA诱导幼鼠哮喘模型的炎症的影响。方法:将40只SD雌性小鼠随机分为正常组、模型组、木犀草素组。小鼠通过第1d、7d腹腔注射卵清蛋白的乳化液,第15d、16d、17d、18d、19d雾化吸入含3%OVA的生理盐水,建立哮喘模型。第15d,LT组每天腹腔注射1mg/kg、2 mg/kgLT;模型组、正常组注射等体积生理盐水。第30d采用小鼠无创肺功能仪检测小鼠气道高反应,ELISA测血清IL-2、IL-4浓度,HE染色观察肺组织炎症变化,RT-PCR检测肺组织TLR4、NF-κB的RNA表达,免疫印迹检测肺组织TLR4/NF-κBpP65蛋白表达。结果:木犀草素组的气道高反应和血清IL-4浓度低于模型组,血清IL-2浓度高于模型组。与模型组比,肺组织中,木犀草素组肺部炎症改善,木犀草素组TLR4、NF-κB的mRNA和蛋白表达低于模型组。结论:木犀草素通过调控TLR4/NF-κB途径抑制幼鼠哮喘模型的炎症反应。
Objective:To investigate the effect of luteolin(LT)on inflammation via the TLR4/NF-κB pathway in an OVA-in⁃duced asthma model in young rats.Methods:40 SD female pups were randomly divided into normal group,model group,and ligno⁃caine group.The asthma model was established in rats by intraperitoneal injection of emulsion of ovalbumin on day 1 and 7,and nebulized inhalation of saline containing 3%OVA on day 15,16,17,18 and 19.On the 15th day,1 mg/kg and 2 mg/kg lignocaine were injected in the lignocaine group,and equal volumes of saline were injected in the model and normal groups,respectively.On the 30th day,the airway hyperresponsiveness of mice was detected by non-invasive lung function instrument,serum IL-2 and IL-4 concentrations were measured by ELISA,inflammatory changes in lung tissues were observed by HE staining,RNA expression of TLR4 and NF-κB in lung tissues was detected by RT-PCR,and TLR4/NF-κB pP65 protein expression in lung tissues was detected by immunoblotting.Results:The airway hyperresponsiveness and serum IL-4 concentrations were lower in the lignocaine group than in the model group,and serum IL-2 concentrations were higher than in the model group.Compared with the model group,in lung tissue,lung inflammation was improved in the lignocaine group,and mRNA and protein expression of TLR4 and NF-κB were lower in the lignocaine group than in the model group.Conclusion:Lignocaine suppresses inflammatory responses in a young mouse asthma model by modulating the TLR4/NF-κB pathway.
