摘要
目的利用非靶向代谢组学方法分析儿童Graves病(Graves′disease,GD)血清代谢组学的改变。方法选取2017年3月~2018年10月首次在苏州大学附属儿童医院内分泌科就诊的青春前期GD患儿30例作为GD组,选取同期在苏州大学附属儿童医院进行正常体检的儿童30例作为对照组。采用超高效液相色谱-四极杆飞行时间质谱法(ultra-high performance liquid chromatography-quadrupole time-of-flight mass spectrometry,UHPLC-QTOF/MS)分析受检者的血清代谢物。结果通过非靶向代谢组学分析,在所有受检者中共鉴定出730个代谢物,共筛选出48个差异代谢物,与对照组比较,GD组有33个代谢物水平升高,15个代谢物水平降低。通路分析显示,GD组中氨基酰-tRNA生物合成、苯丙氨酸代谢、酪氨酸代谢、嘌呤代谢和嘧啶代谢均有显著改变。结论通过基于UHPLC-TOF-MS的非靶向代谢组方法分析发现,GD患儿血清氨基酸代谢及核苷酸代谢组学模式有显著改变。
Objective To analyze the changes of serum metabonomics in children with Graves′disease(GD)based on the untargeted metabonomics techniques.Methods A total of 30 newly diagnosed GD children who were first diagnosed from March 2017 to October 2018 in the Department of Endocrinology,Children′s Hospital of Soochow University were selected as the GD group,and a total of 30 cases of normal healthy children in the Children′s Hospital of Soochow University during the same period were selected as the control group.The serum metabolites of participants were analyzed by ultra-high performance liquid chromatography-quadrupole time-of-flight mass spectrometry(UHPLC-QTOF/MS).Results By untargeted metabonomics analysis,730metabolites were identified in all participants,including 48differential metabolites in the GD group and control group.Compared with the control group,33metabolites were increased and 15metabolites were decreased in the GD group.Pathway analysis showed that aminoacyl-tRNA biosynthesis,phenylalanine metabolism,tyrosine metabolism,purine metabolism and pyrimidine metabolism were significantly changed in the GD group.ConclusionThrough the untargeted metabolomics analysis based on UHPLC-TOF-MS,it was found that serum metabonomics patterns of GD children had significant changes.
