摘要
目的:探讨分析大血藤煎剂对慢性盆腔炎大鼠子宫TGF-β/Smads通路的影响。方法:选择SD雌性大鼠70只,随机分为7个组,分别为对照组、假手术组、模型组、大血藤高剂量组、中剂量组、低剂量组以及阳性药物组。造模给药21d进行检测,检测致炎侧子宫肿胀程度、血清炎症因子表达,并采用RT-qPCR法及Western blot法检测子宫组织TGF-β1、TGF-β3、Smad3、Smad7 mRNA及蛋白表达。结果:大血藤各剂量组及阳性药物组子宫肿胀程度低于模型组(P<0.05),且随着大血藤药物浓度的增加,大鼠右侧子宫肿胀度减轻(P<0.05)。大血藤高、中、低剂量组及阳性药物组大鼠血清IL-6、TNF-α以及子宫TGF-β1、TGF-β3、Smad3、Smad7蛋白和mRNA相对表达量较模型组降低(P<0.05),且随着大血藤药物浓度的增加,各指标降低程度越明显(P<0.05)。结论:大血藤煎剂可有效降低慢性盆腔炎大鼠子宫肿胀,其作用机制可能包括抑制大鼠及炎症反应以及抑制TGF-β/Smads信号通路。
Objective:To investigate and analyze the effects of decoction of Radix hematotensiae on TGF-β/Smads pathway of uterus in rats with chronic pelvic inflammatory disease.Methods:Seventy female SD rats were randomly divided into 7 groups:control group,sham operation group,model group,high dose group,medium dose group,low dose group and positive drug group.21d after the model administration,the serum and uterine tissue of rats were taken to detect the degree of inflammation-induced uterine swelling and the expression of inflammatory factors in serum,and the mRNA and protein expressions of TGF-β1,TGF-β3,Smad3 and Smad7 in uterine tissue were detected by RT-qPCR and Western blot.Results:Compared with the control group and the sham operation group,the right uterus of the rats in the other groups was significantly swollen(P<0.05),while the degree of uterine swelling in the high,medium and low dose groups and the positive drug group of Caulis Dracaena was significantly lower than that in the model group(P<0.05),and with the increase of the concentration of Caulis Dracaena,the degree of uterine swelling on the right side of the rats was significantly reduced(P<0.05).Compared with control group and sham operation group,serum levels of IL-6 and TNF-αin other groups were significantly increased(P<0.05),while serum levels of IL-6 and TNF-αin high-dose,medium-dose and low-dose groups and positive drug groups were significantly decreased compared with model group(P<0.05).Serum IL-6 and TNF-αwere significantly decreased(P<0.05).Compared with control group and sham operation group,the protein and mRNA relative expression levels of TGF-β1,TGF-β3,Smad3 and Smad7 in uterus of other groups were significantly increased(P<0.05).The protein and mRNA relative expression levels of TGF-β1,TGF-β3,Smad3 and Smad7 in uterus of rats in high-dose,medium-dose and low-dose groups and positive drug groups were significantly decreased compared with model group(P<0.05),and with the increase of drug concentration,The protein and mRNA relative
