活血生肌方外用对大鼠压力性损伤组织金属蛋白酶-9及其抑制因子-1表达的影响 被引量:1

Effects of external application of Huoxue Shengji Recipe on expression of metalloproteinase-9 and its inhibitor-1(TIMP-1)in a rat model of pressure-induced tissue injury

摘要 目的观察活血生肌方外用对大鼠压力性损伤组织金属蛋白酶-9(MMP-9)及其抑制因子-1(TIMP-1)表达水平的影响,探讨作用机制。方法60只SD雄性大鼠随机分为治疗组和对照组,各30只,每只大鼠背部建立两个压力性损伤创面。治疗组给予创面常规处理,活血生肌纱条外敷后用无菌纱布包扎,1次/d,直至实验结束。对照组给予创面常规处理,予凡士林纱条外敷后用灭菌纱布包扎,1次/d,直至实验结束。于换药第1、3、5、7、9天,每次换药开始前选取各组6只大鼠切取其背部创面中心部位皮肤组织,采用免疫组化及Masson染色结合图像分析的方法检测大鼠创面组织胶原纤维表达量、Ⅰ型胶原蛋白表达量、MMP-9及TIMP-1表达的变化,将取材后的12只大鼠处死,其余大鼠继续实验。结果换药第3天直至实验结束,治疗组创面组织MMP-9含量较对照组降低[(0.381±0.052)比(0.429±0.061)、(0.308±0.035)比(0.382±0.037)、(0.253±0.022)比(0.328±0.025)、(0.187±0.015)比(0.230±0.021),P均<0.01];TIMP-1含量较对照组升高[(3.118±0.632)比(2.607±0.572)、(4.268±0.518)比(3.351±0.617)、(5.715±0.745)比(4.603±0.685)、(7.250±1.521)比(6.203±1.025),P均<0.05];从换药第5天开始,治疗组创面组织内Ⅰ型胶原蛋白及胶原纤维表达量均高于对照组[Ⅰ型胶原蛋白:(9.8±2.3)比(8.1±1.6)、(13.2±3.8)比(10.3±2.2)、(17.5±4.5)比(13.6±3.2);胶原纤维:(28.8±7.1)比(20.5±6.2)、(37.2±7.5)比(26.6±6.5)、(45.2±8.5)比(32.6±7.9),P均<0.05]。结论活血生肌方可能通过调节压力性损伤组织MMP-9及TIMP-1表达水平,从而起到减少胶原蛋白的降解,促进胶原纤维合成、肉芽组织生长的作用。 Objective To assess the effect of Huoxue Shengji Recipe on expression of metalloproteinase-9(MMP-9)and its inhibitor-1(TIMP-1)in a rat model of pressure-induced tissue injury and then to explore the underlying molecular mechanism of the action.Methods Sixty male Sprague-Dawley(SD)rats were used to establish two stress injury wounds on the back and then randomly divided into control and treatment groups(n=30 per group).The treatment group of rats was treated with conventional wound cleaning and the wound area was wrapped with sterile gauze after external application of Huoxue Shengji gauze strip daily for 9 days.In contrast,the control rats were just treated with a routine wound cleaning and the wound area was applied with Vaseline gauze daily for9 days.After that,on the 1st,3rd,5th,7th,and 9th day before the daily treatment,6 rats in each group were randomly selected to take a biopsy of the skin at the central wound area for immunohistochemistry and Masson staining combined with image analysis to detect changes in Collagen fiber,collagen type I,MMP-9 and TIMP-1expression.These 12 rats in each time point were sacrificed and the rest rats were continued for the experiment.Results On the third day of treatment until end of the experiment,level of MMP-9 was lower in the wound tissues of the treatment rats than that of the control rats(0.381±0.052 vs.0.429±0.061;0.308±0.035 vs.0.382±0.037;0.253±0.022 vs.0.328±0.025;0.187±0.015 vs.0.230±0.021;P<0.01),while TIMP-1 level increased(3.118±0.632vs.2.607±0.572;4.268±0.518 vs.3.351±0.617;5.715±0.745 vs.4.603±0.685;7.250±1.521 vs.6.203±1.025;P<0.05).Moreover,level of type I collagen and collagen fiber was higher in the wound tissues of the treatment rats than those of the control rats after 5 day-treatment(9.8±2.3 vs.8.1±1.6;13.2±3.8 vs.10.3±2.2;17.5±4.5 vs.13.6±3.2 and 28.8±7.1 vs.20.5±6.2;37.2±7.5 vs.26.6±6.5;45.2±8.5 vs.32.6±7.9,respectively;all P<0.05).Conclusion The Huoxue Shengji Recipe was able to reduce collagen degradation,but promote colla
