己糖激酶Ⅱ在乳腺癌中的表达及对细胞增殖能力的影响

摘要 目的探讨HKⅡ在浸润性乳腺导管癌(invasive ductal carcinomas,IDC)中的表达和临床意义及HKⅡ对乳腺癌细胞增殖能力的影响。方法通过免疫组织化学法研究IDC组织和癌旁正常组织中HKⅡ的表达水平;使用Spearman检验分析HKⅡ的表达与浸润性乳腺导管癌患者临床特征之间的相关性;使用Kaplan-Meier方法和Log-rank检验评估浸润性乳腺导管癌中HKⅡ的表达水平与总生存期之间的关系;使用Cox比例风险回归模型进一步评估HKⅡ表达水平对浸润性乳腺导管癌患者总生存期的预测效能。通过瞬时转染法将体外培养的MCF 7和MDA-MB-231细胞进行HKⅡ干扰处理,Western Blot法检测HKⅡ蛋白的表达水平;葡萄糖氧化酶法检测培养液中葡萄糖浓度;MTT法检测细胞增殖情况。结果69例IDC组织中HKⅡ的表达水平显著高于相应正常乳腺组织;138例IDC组织中HKⅡ高表达的IDC患者生存期比HKⅡ低表达患者更短,而且HKⅡ高表达是IDC患者预后不良的独立危险因素;KMPLOT数据库的3951位乳腺癌患者HKⅡ的表达水平与其生存期同样存在负相关。干扰HKⅡ后消耗的葡萄糖减少,细胞增殖速度减慢。结论HKⅡ在IDC患者中的表达明显高于正常乳腺组织,HKⅡ高表达是IDC患者预后不良的独立危险因素;而且HKⅡ促进乳腺癌细胞的葡萄糖的摄取及细胞增殖。 Objective To investigate the expression and clinical significance of HKⅡin invasive ductal carcinoma(IDC)and the effect of HKⅡon the proliferation of breast cancer cells.Methods Immunohistochemistry was performed to investigate the expression of HKⅡin IDC tissues and adjacent normal tissues.Spearman test was used to analyze the correlation between the expression of HKⅡand the clinical characteristics of invasive breast ductal carcinoma.Kaplan-Meier method and Log-rank test were used to evaluate the association between HKⅡexpression and overall survival in invasive ductal carcinoma of the breast.Cox proportional hazards regression model was used to further evaluate the predictive power of HKⅡexpression level on overall survival in patients with invasive ductal carcinoma.MCF 7 and MDA-MB-231 cells cultured in vitro were interfered with HKⅡby transient transfection method,and the expression of HKⅡprotein was detected by Western Blot.Glucose oxidase method was used to detect the concentration of glucose in culture medium.Cell proliferation was detected by MTT assay.Results The expression level of HKⅡin 69 cases of IDC tissues was significantly higher than that in corresponding normal breast tissues;the survival time of 138 IDC patients with high expression of HKⅡwas shorter than that of patients with low expression of HKⅡ,and high expression of HKⅡwas an independent risk factor for poor prognosis of IDC patients;the expression level of HKⅡwas also negatively correlated with the survival time of 3,951 breast cancer patients in KMPLOT database.After interfering with HKⅡ,the consumption of glucose decreased and the cell proliferation slowed down.Conclusion The expression of HKⅡin IDC patients was significantly higher than that in normal breast tissue.High expression of HKⅡis an independent risk factor for poor prognosis in IDC patients;HKⅡpromoted glucose uptake and cell proliferation in breast cancer cells.
