基于JAK1/STAT3信号通路探讨寿胎丸含药血清对人绒毛膜滋养层细胞增殖、侵袭和迁移的影响 被引量:8

摘要 目的观察寿胎丸含药血清对人绒毛膜滋养层细胞HTR-8/Svneo增殖、侵袭和迁移的影响,基于Janus激酶1(JAK1)/信号转导和转录激活因子3(STAT3)通路探讨其作用机制。方法制备寿胎丸含药血清及空白血清,体外培养HTR-8/Svneo细胞,将细胞分为对照组和寿胎丸低、中、高剂量组,分别用空白血清及2.5%、5%、10%含药血清处理细胞,CCK8法检测细胞存活率,划痕实验、Transwell实验检测细胞迁移和侵袭能力,Western blot检测HTR-8/Svneo细胞JAK1/STAT3通路相关蛋白及增殖相关蛋白Ki67、血管内皮细胞生长因子受体(VEGFR)-2、基质金属蛋白酶(MMP)-9、微染色体维持蛋白2(MCM2)、增殖核抗原(PCNA)表达,RT-PCR检测Ki67、PCNA mRNA表达。结果与对照组比较,寿胎丸低、中、高剂量组细胞存活率显著升高,且作用36 h细胞存活率最高(P<0.05,P<0.01),寿胎丸中、高剂量组细胞划痕愈合率显著升高(P<0.05,P<0.01),寿胎丸低、中、高剂量组细胞迁移率及侵袭率显著升高(P<0.01),寿胎丸高剂量组细胞p-JAK1、p-STAT3蛋白表达显著升高(P<0.05),寿胎丸中、高剂量组细胞Ki67、VEGFR-2、MMP-9、MCM2、PCNA蛋白表达显著升高(P<0.05,P<0.01),寿胎丸低、中、高剂量组细胞Ki67、PCNA mRNA表达显著升高(P<0.05,P<0.01),以寿胎丸高剂量组作用最显著(P<0.05,P<0.01)。结论寿胎丸含药血清能提高HTR-8/Svneo细胞增殖、侵袭和迁移能力,其机制可能与激活JAK1/STAT3信号通路有关。 Objective To observe the effects of Shoutai Pills-containing serum on the proliferation,invasion and migration of human chorionic trophoblast cells(HTR-8/Svneo);To explore its mechanism based on the JAK1/STAT3 pathway.Methods Shoutai Pills-containing serum and blank serum were prepared,HTR-8/Svneo cells were cultured in vitro,and the cells were divided into control group(treated with blank serum)and Shoutai Pills low-,medium-and high-dosage groups(treated with drug-containing serum concentration 2.5%,5%,10%).CCK8 method was used to detect cell viability,scratch test and Transwell test were used to detect cell migration and invasion ability,Western blot was used to detect the expression of JAK1/STAT3 pathway-related proteins,cell proliferation-related proteins Ki67,VEGFR-2,MMP-9,MCM2,and PCNA in HTR-8/Svneo cells.RT-PCR was used to detect Ki67,PCNA mRNA expression.Results Compared with the control group,the cell viability significantly increased in Shoutai Pills low-,medium-and high-dosage groups,and the cell viability was the highest at 36 h(P<0.05,P<0.01),the scratch healing rate increased significantly in Shoutai Pills medium-and high-dosage groups(P<0.05,P<0.01),the migration rate and invasion rate significantly increased in Shoutai Pills low-,medium-and high-dosage groups(P<0.01),the proteins expression of p-JAK1 and p-STAT3 in cells significantly increased in Shoutai Pills high-dosage group(P<0.05),the proteins expression of Ki67,VEGFR-2,MMP-9,MCM2 and PCNA significantly increased in Shoutai Pills,medium-and high-dosage groups(P<0.05,P<0.01),the expressions of Ki67 and PCNA mRNA significantly increased in Shoutai Pills low-,medium-and high-dosage groups(P<0.05,P<0.01),and the Shoutai Pills high-dosage group had the most significant effect(P<0.05,P<0.01).Conclusion Shoutai Pills-containing serum can promote the proliferation,invasion and migration of HTR-8/Svneo cells,and the mechanism may be related to the activation of JAK1/STAT3 signaling pathway.
