摘要
本研究建立了一种柱前衍生-离子液体微萃取-高效液相色谱方法,用于发酵食品中3种生物胺的分析检测。方法采用荧光胺作为生物胺衍生试剂,并以疏水性1-辛基-3-甲基咪唑六氟磷酸盐作为萃取剂,以亲水性的1-己基-3-甲基咪唑四氟硼酸盐作为分散剂,进行生物胺衍生物的萃取,并对萃取剂种类和用量,分散剂用量,萃取温度等进行优化。在优化实验条件下,3种生物胺衍生物在1~100 mg/kg范围内呈现良好的线性关系,相关系数为0.9964~0.9993,检出限为0.39~0.91 mg/kg,定量限为1.52~5.36 mg/kg;实际发酵样品的加标回收率在81.0%~108.7%范围内,相对标准偏差(RSD)小于6%。该方法成功用于小米辣和腌菜等发酵食品中生物胺的测定。
In this study,a pre-column derivatization-ionic liquid microextraction-high performance liquid chromatography method was established and used for the analysis of three biogenic amines in fermented foods.Fluorescamine,hydrophobic 1-octyl-3-methylimidazolium hexafluorophosphate and hydrophilic 1-hexyl-3-methylimidazolium tetrafluoroborate were used as biogenic amine derivatizing reagents,extractants and dispersants,respectively,for the extraction of biogenic amine derivatives.The type and dosage of extractant,dosage of dispersant,and extraction temperature were optimized.The three biogenic amine derivatives showed a good linear relationship in the range of 1-100 mg/kg,the correlation coefficient was 0.9964-0.9993,the detection limit was 0.39-0.91 mg/kg,and the quantification limit was 1.52-5.36 mg/kg.The spiked recoveries of the fermentation samples were in the range of 81.0%-108.7%,and the relative standard deviation was less than 6%.The method was successfully used for the determination of biogenic amines in fermented foods including millet spicy and pickled vegetables.
