摘要
目的:基于网络药理学研究方法探讨乌梅丸治疗抑郁症的可能作用机制。方法:应用中药系统药理学数据库与分析平台(TCMSP)、Uniprot数据库、PubChem数据库、Swiss Target Prediction数据库筛选乌梅丸的生物活性成分,并预测其靶点;应用OMIM、Genecards、TTD数据库收集抑郁症相关靶点;利用Venny 2.1软件对乌梅丸与抑郁症的靶点进行映射,获得药物-疾病共同靶点,并绘制韦恩图,构建“药物-成分-靶点-疾病”网络图,筛选获得核心活性成分;运用STRING数据库及Cytoscape 3.7.2软件构建蛋白质相互作用(PPI)网络,筛选获得核心靶点;应用Bioconductor生物信息软件包,进行药物-疾病共有靶基因基因本体(GO)功能注释和京都基因与基因组百科全书(KEGG)通路富集分析。结果:预测得到乌梅丸治疗抑郁症的重要活性成分39种,核心靶点18个,GO富集分析结果为生物学过程1577个、细胞组分112个、分子功能148个,KEGG富集分析得到信号通路132条。其中,人参皂苷Rg5、人参二醇、五味子酯乙、黄藤素、山柰酚、槲皮素、β-谷甾醇可能是乌梅丸治疗抑郁症的重要活性成分,潜在核心靶点为AKT1、APP、MAPK3、MAPK1、CXCL8等,KEGG通路富集分析主要涉及神经发生、突触可塑性、程序性细胞死亡、炎症免疫等多方面机制变化。结论:乌梅丸在抑郁症治疗中具有多成分-多靶点-多途径的特点与优势,此可作为进一步探讨乌梅丸治疗抑郁症的药效物质及作用机制重要的研究基础。
Objective:To explore the possible mechanism of Wumei Pills in the treatment of depression based on the research method of network pharmacology.Methods:Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform(TCMSP),Uniprot database,PubChem database and Swiss Target Prediction database were used to screen the biologically active ingredients of Wumei Pills and to predict their targets;OMIM,Genecards and TTD databases were used to collect depression-related targets;Venny 2.1 software was used to map the targets of Wumei Pills and depression,to obtain common drug-disease targets and draw a Venn diagram,to construct a‘drug-component-target-disease’network diagram,and to screen the key active ingredients;STRING database and Cytoscape 3.7.2 software were used to construct a protein-protein interaction(PPI)network and to screen the core targets;Bioconductor biological information software package was used to perform gene ontology(GO)annotations and Kyoto encyclopedia of genes and genomes(KEGG)pathway enrichment analyses for the drug-disease shared target genes.Results:It was predicted that,in the treatment of depression,Wumei Pills obtained 39 key active ingredients and 18 core targets.The result of GO annotations consisted 1577 biological processes,112 cell components,and 148 molecular functions.The result of KEGG enrichment analyses consisted 132 signal pathways.Among them,ginsenoside Rg5,panaxadiol,gomisin B,palmatine,kaempferol,quercetin,β-sitosterol may be the key active ingredients of Wumei Pills in the treatment of depression,and the potential core targets may include AKT1,APP,MAPK3,MAPK1,CXCL8,etc.Enrichment analyses of KEGG pathway mainly involved the changes in many aspects,such as neurogenesis,synaptic plasticity,programmed cell death,inflammatory immunity,and so on.Conclusion:Wumei Pills has the characteristics and advantages of‘multi-component,multi-target,multi-pathway’in the treatment of depression,which can be used as an important research basis for further exploring the p
