摘要
目的:探讨枸杞多糖(LBP)调控Wnt信号通路对子宫肌瘤细胞生物活性的影响。方法:将健康的30只SD大鼠随机分为5组:对照组、模型组、LBP低剂量组、LBP中剂量组和LBP高剂量组,每组6只,造模结束后,低剂量组,中剂量组和高剂量组大鼠分别给予10、50和100 mg/kg LBP灌胃,1次/d,连续14 d,对照组和模型组大鼠相同时间内给予相同剂量的生理盐水灌胃,检测子宫系数。选择10例子宫肌瘤组织进行分离,子宫肌瘤细胞培养至密度为5×10^(7)个/ml,将细胞随机分为4组,子宫肌瘤组、低LBP组、中LBP组和高LBP组,后3组子宫肌瘤细胞加入100、200、400μg/ml的LBP,连续干预72 h,采用MTT检测、流式细胞仪、Transwell小室分别检测子宫肌瘤细胞存活率、凋亡率及侵袭数目;采用Western blot及qRT-PCR检测Wnt/β-catenin蛋白及mRNA表达。结果:与对照组相比,模型组大鼠的体质量降低,子宫重量、子宫系数明显提高(P<0.05),与模型组相比,LBP低、中及高剂量组大鼠子宫重量、子宫系数均降低,体质量增加,组间比较差异均有统计学意义(P<0.05);CCK-8检测结果显示:低、中、高LBP组24 h、48 h及72 h组子宫肌瘤存活率均低于子宫肌瘤组(均P<0.05),子宫肌瘤组在24 h、48 h及72 h时差异均无统计学意义(P>0.05),低、中、高LBP组在24 h时细胞存活率差异无统计学意义(P>0.05),其余时间点两两比较子宫肌瘤存活率差异均有统计学意义(均P<0.05);流式细胞仪结果显示:低、中、高LBP组子宫肌瘤凋亡率均高于子宫肌瘤组(均P<0.05),低、中、高LBP组两两相比子宫肌瘤凋亡率差异均有统计学意义(均P<0.05);Transwell小室结果显示:低、中、高LBP组子宫肌瘤细胞侵袭数目均低于子宫肌瘤组(均P<0.05),低、中、高LBP组两两相比子宫肌瘤细胞侵袭数目差异具有统计学意义(均P<0.05);子宫肌瘤组细胞中Wnt5b和β-catenin蛋白/mRNA表达水平低于低、中及高LBP组(均P<0.05),低、中�
Objective:To investigate the biological activity and Wnt signaling pathway of lycium barbarum polysaccharide(LBP)on uterine fibroids.Methods:Thirty healthy SD rats were randomly divided into 5 groups,control group,model group,LBP low-dose group,LBP medium-dose group and high-dose group,with 6 in each group,rats in the LBP low-dose group,LBP mediumdose group and LBP high-dose group were given 10,50 and 100 mg/kg LBP by gavage after modeling,once a day,for 14 consecutive days.Rats in the control group and the model group were given the same dose of normal saline by gavage at the same time,and the uterine coefficient was measured.Ten cases of uterine leiomyoma tissue were isolated and cultured to a cell density of 5×10^(7)cells/ml.The cells were randomly divided into 4 groups:uterine leiomyoma group,low LBP group,medium LBP group and high LBP group.The cells in the latter three groups were treated with 100,200,400μg/ml LBP for 72 h.MTT assay,flow cytometry and Transwell chamber were used to detect the survival rate,apoptosis rate and invasion number of uterine leiomyoma cells after 72 h of intervence.Western blot and RT-PCR were used to detect the protein and mRNA expression of Wnt/β-catenin.Results:Compared with control group,the model group had a significant reduction in body weight and a significant increase in uterine weight and uterine coefficient(P<0.05).Compared with the model group,the LBP low-dose,medium-dose and high-dose groups had a significant reduction in uterine weight and uterine coefficient and a significant increase in body weight(P<0.05).CCK-8 results showed that the survival rate of uterine fibroids in the low,medium and high LBP groups at 24 h,48 h and 72 h was lower than that in the uterine fibroid group(all P<0.05),and there was no significant difference in the uterine fibroid group at 24 h,48 h and 72 h(P>0.05).There was no significant difference in the cell survival rate among the low,medium and high LBP groups at 24 h(P>0.05),but there were significant differences in the survival rate of
