基于胃癌关键基因的中药抗肿瘤研究现状分析 被引量:3

Research Status of Key Genes-target Chinese Herbal Medicine for the Treatment of Gastric Cancer

摘要 目的分析以胃癌关键基因为治疗靶点的中药在胃癌防治中的研究现状。方法在GEO数据库中下载胃癌基因表达谱芯片数据,经筛选后确定GSE54129和GSE64916两个数据集,以“P<0.05,Log2FC≥1”为截断值进行肿瘤组织与正常组织的差异表达基因筛选,利用韦恩图取两个数据集的交集,进行基因本体论(GO)分析、京都基因与基因组百科全书(KEGG)通路富集分析及差异表达基因的蛋白互作(PPI)网络分析,采用Cytoscape(3.8.2版本)软件计算得出的关键基因,在Coremine Medical数据库中对关键基因进行检索,在“traditional Chinese medicine”条目中得到以关键基因作为治疗靶点的中药。对得到的中药进行频次统计,选择前5味中药,在中文及外文数据库中对其在胃癌中的相关研究进行文献检索。将检索到的文献分为临床研究、网络药理学分析、数据挖掘/处方经验、基础实验,对研究现状进行总结。结果韦恩图取交集后得到两个数据集有150个共同的差异表达基因,富集到的生物功能与通路主要有PI3K/AKT通路、上皮细胞转移、胶原蛋白细胞外基质等。利用String绘制PPI网络,进一步在Cytoscape中根据自由度大小选出前10位的关键基因:CDKN2A、SPP1、CTGF、C3、CST3、APOB、GC、IL18、CCND1、CD34。应用Coremine medical数据库分析得到治疗关键基因靶点的中药,对检索到的中药进行频次统计,出现频次最高的前五味中药分别为黄芪(与9个基因有关),白花蛇舌草、淫羊藿、三七(各与4个基因有关),菝葜(与3个基因有关)。外文数据库未检索到相关文献,于2022年2月20日在中文CNKI数据库分别以“胃癌”“胃恶性肿瘤”and“黄芪”“白花蛇舌草”“淫羊藿”“三七”“菝葜”进行检索,仅中药黄芪有临床研究,共49篇;网络药理学分析中黄芪有8篇,白花蛇舌草3篇,其他中药未检索到;数据挖掘/处方规律研究中黄芪21篇,白花蛇舌草4篇,� Objective To analyze the research status of gastric cancer(GC)key genes-targeted Chinese herbal medicinals(CHMs)in the prevention and treatment of GC.Methods The chip data of GC gene expression profiling was downloaded from GEO database.After screening,GSE54129 and GSE64916 data sets were selected.The diffe-rentially expressed genes between tumor and normal tissues were screened out with“P<0.05,log2fc≥1”as the cut-off value.Wayne diagram was used to take the intersection of the two data sets for Gene Ontology(GO)analysis,Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway enrichment and protein-protein interaction(PPI)analysis.The key genes calculated by Cytoscape software 3.8.2 were searched in the Coremine Medical database,and the key genes-targeted CHMs were obtained in the entry of“traditional Chinese medicine”.The frequency of the obtained CHMs were counted,and the top five CHMs were searched in the literature databases for their related researches on GC.The retrieved literature was classified into four categories including clinical research,network pharmacology analysis,data mining/prescription experience,and experimental research,and the research status was summarized.Results The two data sets obtained through intersection by Wayne diagram had 150 common differentially expressed genes.The enriched biological functions and pathways mainly included PI3K/AKT pathway,epithelial cell transfer,and collagen extracellular matrix.Using String to draw PPI network,the top 10 key genes obtained in Cytoscape according to the degree of freedom were CDKN2A,SPP1,CTGF,C3,CST3,apoB,GC,IL18,CCND1 and CD34.The Coremine medical database was used to analyze the key genes-target CHMs,and the top five CHMs with the highest frequency were Huangqi(Radix Astragali)in relation to 9 genes,Baihua Sheshe Cao(Herba Hedyotis Diffusae),Yinyanghuo(Herba Epimedii),and Sanqi(Radix et Rhizoma Notoginseng)each in relation to four genes,and Baqia(Rhizoma Smilacis Chinae)in relation to three genes.No relevant literature was found in th
