摘要
目的 探究颈通康对重度颈动脉狭窄模型大鼠的影响及所涉及的信号通路机制。方法 将(200±20)g的SD雄性大鼠36只按随机数字表随机分为模型组、假手术组、补阳还五汤组(即原方组)、颈通康组(即加味方组),每组9只。假手术组给予普通饲料喂养,颈部切开但不进行颈总动脉钳夹术,模型组、原方组、颈通康组均采用颈动脉钳夹联合高脂饮食、维生素D3注射构建颈动脉粥样硬化性狭窄大鼠模型。造模成功后,原方组予补阳还五汤原方以13.3 mg/kg剂量灌胃给药,颈通康组予颈通康以13.3 mg/kg剂量灌胃给药。假手术组与模型组则用等量生理盐水灌胃,各组均连续给药4周。干预结束后,应用qRT-PCR分析模型大鼠颈动脉组织中PI3K、AKT、NF-κB-p65 mRNA蛋白表达情况;应用Western Blot分析模型大鼠颈动脉组织中PI3K、AKT、NF-κB表达情况。结果 经Western Blot检测分析,与假手术组比较,模型组、原方组、颈通康组大鼠PI3K、AKT、NF-κB(p65)蛋白表达显著升高(P<0.01);与模型组比较,原方组、颈通康组大鼠PI3K、AKT、NF-κB(p65)蛋白表达显著降低(P<0.01);与原方组比较,颈通康组大鼠PI3K、AKT、NF-κB(p65)蛋白表达显著降低(P<0.01)。经qRT-PCR检测分析,与假手术组比较,模型组、原方组大鼠PI3K、AKT、NF-κB mRNA表达显著升高(P<0.01),颈通康组PI3K、NF-κB mRNA表达显著升高(P<0.01)而AKT mRNA表达差异无统计学意义;与模型组比较,原方组、颈通康组大鼠PI3K、AKT、NF-κB mRNA表达显著降低(P<0.01);与原方组比较,颈通康组大鼠,AKT mRNA表达显著降低(P<0.01)而PI3K、NF-κB mRNA表达差异无统计学意义。结论 补阳还五汤、颈通康至少能够通过抑制PI3K/AKT/NF-κB信号通路减轻大鼠颈动脉狭窄。其中,颈通康疗效优于补阳还五汤原方。
Objective To explore the effect of Jingtongkang(颈通康)on severe carotid artery stenosis model rats and the involved signaling pathway mechanism.Methods Thirty-six(200±20)g SD male rats were randomly divided into model group,sham-operated group,Buyang Huanwu Decoction(补阳还五汤)group(original formula group)and Jingtongkang group by the random number table,9 in each group.The sham operation group was given ordinary feeding and the neck was cut open,but common carotid artery clamp was not performed.The model group,the original prescription group and the Jingtongkang group were all used carotid artery clamp combined with high-fat diet and vitamin D3 injection to construct a rat model of atherosclerotic carotid stenosis.After successful modeling,the original prescription group was given original Buyang Huanwu Decoction by gavage at a dose of 13.3 mg/kg,and the Jingtongkang group was given Jingtongkang by gavage at a dose of 13.3 mg/kg.The sham operation group and the model group were given the equal amount of normal saline by intragastric administration and each group was continuously administered for four weeks.After the intervention,qRT-PCR was used to analyze the protein expressions of PI3 K,AKT and NF-κB-p65 mRNA in the carotid artery tissue of the model rats.Western Blot was used to analyze the expression of PI3 K,AKT and NF-κB in the carotid artery tissue of the model rats.Results After Western Blot detection and analysis,compared with those of the sham operation group,the expressions of PI3 K,AKT and NF-κB(p65)proteins in the model group,the original prescription group and the Jingtongkang group were obviously increased(P<0.01).Compared with those of the model group,the expressions of PI3 K,AKT and NF-κB(p65)proteins in the original prescription group and the Jingtongkang group were obviously reduced(P<0.01).Compared with those of the original prescription group,the expressions of PI3 K,AKT and NF-κ(p65)in the Jingtongkang groupn was obviously reduced(P<0.01).After qRT-PCR detection and analysis
