摘要
目的建立无水乙醇致大鼠急性胃溃疡模型,探讨清热愈溃混悬剂对实验性胃溃疡大鼠的保护作用。方法选取体质量均匀的SD大鼠随机分为正常对照组、模型对照组、阳性对照组与清热愈溃混悬剂低、中、高剂量组,每组8只。建立无水乙醇致大鼠实验性胃溃疡模型,评价口服给予清热愈溃混悬剂低、中、高剂量〔2.2、3.3、4.4 g·(kg·d)^(-1)〕对大鼠实验性胃溃疡的预防效果。结果与正常对照组比较,模型对照组大鼠胃黏膜表面出现大量淤血,上皮细胞脱落坏死,清热愈溃混悬剂可使大鼠胃黏膜溃疡面积显著减小;可明显降低大鼠胃组织中丙二醛、白介素-1β、白介素-6与肿瘤坏死因子-α水平(P<0.01);可升高胃组织中谷胱甘肽、超氧化物歧化酶与表皮生长因子水平(P<0.01)。结论清热愈溃混悬剂可通过抗氧化应激、抗炎症等作用对大鼠胃黏膜产生保护作用。
Objective To establish a model of acute gastric ulcer induced by anhydrous ethanol and to investigate the protective effect of Qingreyukui suspension.Methods SD rats of uniform body weight were randomly divided into the normal control group,model control group,positive control group〔Lansoprazole,30 mg·(kg·d)^(-1)〕,Qingreyukui suspension low-dose group,medium-dose group and high-dose group,with 8 rats in each.An experimental gastric ulcer model of rats was established via intragastric administration of 5 ml·kg^(-1)absolute ethanol.The preventive effect of Qingreyukui suspension at low,medium and high doses〔2.2,3.3,4.4 g·(kg·d)(-1)〕on experimental gastric ulcer in rats was evaluated.Results Compared with the normal group,the mucosal surface of rats in the model group showed a large amount of congestion,epithelial cells were exocytic and necrotic,and the mucosal ulcer area of these rats was significantly reduced.The levels of malondialdehyde,interleukin-1β,interleukin-6 and tumor necrosis factor-αin gastric tissues of rats were significantly decreased(P<0.01),while glutathione,superoxide dismutase and epidermal growth factors in gastric tissues were increased(P<0.01).Conclusion Qingreyukui suspension can significantly protect gastric mucosa in gastric ulcer rats by combating oxidative stress and inflammation.
