矢车菊素-3-O-葡萄糖苷通过Nrf2/ARE信号通路对蛛网膜下腔出血小鼠血脑屏障完整性影响研究

Effect of cyanidin-3-O-glucoside on blood-brain barrier integrity via Nrf2/ARE signaling pathway in mice with subarachnoid hemorrhage

摘要 目的探讨矢车菊素-3-O-葡萄糖苷(C3G)对蛛网膜下腔出血(SAH)后早期脑损伤(EBI)的保护作用。方法选取54只成年雄性C57BL/6J小鼠随机分成假手术组、模型组及C3G组,每组各18只。模型组采用模型穿刺法建立小鼠SAH模型。建模24 h后,评估小鼠SAH分级评分、加西亚评分,检测小鼠脑水含量以及依文思蓝渗出量。应用蛋白质免疫印迹实验检测小鼠Nrf2,HO-1,NQO1,ZO-1,Occludin及Claudin-5水平。结果建模后24 h,与假手术组相比,模型组、C3G组SAH分级评分、脑含水量及伊文斯蓝渗出量均显著增加,但与模型组比较,C3G组SAH分级评分、脑含水量及伊文斯蓝渗出量均显著降低,差异均有统计学意义(P<0.05)。建模后24 h,与假手术组比较,模型组、C3G组脑组织中Nrf2/ARE信号通路中的分子Nrf2、HO-1、NQO1、ZO-1、Claudin-5及Occludin蛋白表达水平显著降低,但与模型组比较,C3G组Nrf2、HO-1、NQO1、ZO-1、Claudin-5及Occludin蛋白表达水平显著增加,差异有统计学意义(P<0.05)。结论C3G能够作用于Nrf2/ARE信号通路,保护SAH后血脑屏障的完整性。 Objective To investigate the protective effect of cyanidin-3-O-glucoside(C3G)on early brain injury(EBI)after subarachnoid hemorrhage(SAH).Methods Fifty-four adult male C57BL/6J mice were randomly divided into sham operation group(sham group),model group and C3G treatment group(C3G group),with 18 mice in each group.Model group was established by model puncture.After 24 hours of modeling,the SAH grade score and Garcia score of mice were evaluated,and cerebral water content,and Evans blue content of mice were detected.The levels of Nrf2,HO-1,NQO1,ZO-1,Occludin and Claudin-5 were measured by western blot.Results At 24 h after modeling,compared with the sham group,the SAH grade score,brain water content and Evans blue content in the model group and C3G group were significantly increased,but compared with the model group,the SAH grade score,brain water content and Evans blue content in the C3G group were significantly decreased,and the differences were statistically significant(P<0.05).Twenty-four hours after modeling,compared with the sham group,the expression levels of Nrf2,HO-1,NQO1,ZO-1,Claudin-5 and Occludin in the Nrf2/ARE signaling pathway in the brain tissue of the model group and C3G group were significantly decreased,but compared with the model group,the protein expression levels of Nrf2,HO-1,NQO1,ZO-1,Claudin-5 and Occludin in C3G group were significantly increased,and the differences were statistically significant(P<0.05).Conclusion C3G can act on Nrf2/ARE signaling pathway and protect the integrity of blood-brain barrier after SAH.
